Genomic Research LaboratoryTo Setup Genomic Research laboratory to establish state of the art automated testing in Blood Centers for genotype matching. • Used emerging technologies to develop a new program in Transfusion Medicine for genomic testing for red cell antigens. • Created a tech-friendly platform that can effectively type 23 blood groups (100s of alleles, 1000s of SNPs) in one assay (patent pending). • Designed array technology and automation strategies with Information Technology staff to provide automation and effective readout of genomic information. • With funding from United States Armed forces, identified combat ready blood group testing strategy for transfusion safety. Developed systems that ensured regulatory compliance and patient confidentiality in testing systems. Genetic Markers Laboratory• Set up Immunogenetics Typing Laboratory at WaNPRC to identify MHC antigens in nonhuman primate species and develop nonhuman primate transplantation models • Train and direct technical personnel and manage the laboratory • Interact with collaborators, animal technicians, veterinarians, pathologists, and surgeons • Generate nucleotide sequence information from primate species • Design allele discrimination probes for high throughput and high resolution HLA typing • Developed transplantation models to test novel immunosuppression protocols and reverse diabetes induced hyperglycemia by intraportal islet transplantation Involved in two projects, “In utero stem cell transplantation” (funded by NIH) and “Simian Vaccine Evaluation Units”(funded by NIAID). Both were multi-center collaborations that required Animal use and care protocols. Co-investigator for multi-center SEARCH study of Diabetes in Youth (funded by Centers for Disease Control, National Institute of Diabetes and Digestive and Kidney Diseases). Required manipulation of huge sets of data (3700 DNA samples, tested for three loci (25 to 300 alleles for each of these loci). Technology included OneLambda’s Luminex platform and Applied Biosystems 16 channel sequencer ware for genotyping. Thousands of samples processed and data analyzed with patient confidentiality procedures in place• Co-investigator of a mutli-center, international study on diabetes keto-acidosis. Results from these studies were published in National and International journals.

Director, Molecular Biology Laboratory – Division of Research (1996 – 2004) • Identification of disease association markers, inducing donor specific tolerance to bypass immunosuppression, disease association markers, autoimmunie diabetes. • Collaborated with national and international investigators on Chlamydia, HIV, diabetes and transplantation research• Conducted research in transplantation tolerance and therapeutic applications for diabetes• Developed transplantation models to test novel immunosuppression protocols and reverse diabetes induced hyperglycemia by intraportal islet transplantationDirector and Technical consultant – Molecular Biology Laboratory, HLA (1991 – 1995) • Developed molecular typing for HLA to replace traditional serological typing and trained technologists transition to an entirely new way of testing HLA, i.e., from serology to genomics • As the director of molecular biology laboratory for HLA, initiated and managed state-of-the-art sequencing and genotyping laboratories and provided high resolution contract typing for various laboratories in the USA, Canada, Kenya and Europe

• Trained in novel molecular methods, including enzymatic amplification now commonly known as PCR (this experience later led to the identification of nearly 300 major histocompatibility alleles in human and nonhuman primates)

• Joined the Primate Center as Fulbright pre-PhD fellow and worked on developing reagents to type nonhuman primate MHC• Upon successful completion of PhD program with the Panjab University continued as a Research Scientist at the Regional Primate Research Center.• Worked under the guidance of Dr. EA Clark, Professor, Dept Immunology, University of Washington and obtained funding from NCRR to set up Genetic Markers lab. The funding for this laboratory continued for 25 years, that is until May 2007. • Developed MHC typing for macaques• Introduced molecular typing methods

• Sabbatical at Max Plank Institut Für Biologie, in Prof. Jan Klein’s laboratory• Trained in molecular biology, cloning and nucleotide sequencing methods • Screened chimpanzee cDNA library for class II alleles using human class II locus-specific probes for insight into the evolutionary origin of the MHC class II polymorphisms

• Introduced to basic and clinical immunology under the tutelage of Dr. Hugh Fudenberg, Philip Arnaud and Subbi Mathur.
