As a chief investigator, I led a number of research projects to decipher the biological functions of an unusual metamorphic ion channel protein CLIC1. By observing the growth, development and ageing of CLIC1-gene knockout mice in comparison with their wildtype control 129X1SVJ, CLIC1 KO mice were about 10% lighter in body weight and more prone to infections during development, however the survivors had a longer lifespan. This might be related to a reduced ability of CLIC1 KO mice to generate reactive oxygen species.By immunofluorescence confocal microscopy, CLIC1 was found to co-localize with a number of cytoskeletal proteins EMR, and endocytic proteins such as RhoA. By live cell immunofluorescence microscopy, CLIC1 was found to regulate endosomal acidification of macrophages and dendritic cells which were primary cells isolated CLIC1 KO mice.
