Our approach will be to use the pea line PI271119 as a candidate for our investigation, a highly pigmented variety that has been previously transformed within the Dhingra labororatory. Regeneration and genetic transformation will be completed through CRISPR-Cas9 gene knockout technology in order to mutate Flavanone-3-hydroxylase gene, which is involved in the anthocyanin pathway and can be silenced without affecting the overall development of the pea plant. We will screen the transformed pea plant to ensure accurate transformation and use Sanger Sequencing to search for mutations in the Flavanone-3-hydroxylase gene, in order to evaluate the validity of our hypothesis.
