Activity dependent bulk endocytosis (ADBE) occurs at neuronal synapses during high activity to compensate for large amounts of internal membrane that fuse with the plasma membrane. ADBE is more rapid than constitutive clathrin mediated endocytosis and is triggered by high activity. New synaptic vesicles (SVs) derived from the bulk endosomes (BEs) primarily refill the recyclable pool of vesicles yet the intervening steps from BEs to SVs are unknown. Also unclear is whether internalized BEs are distinct from early endosomes where internalized receptors and cargo are sorted. In order to identify the proteins localised to BEs as a first step towards understanding ADBE regulation I developed a purification method to isolate BEs from SVs for subsequent mass spectrometry (MS) analysis. This involved loading the BEs in active synaptosomes with an uptake marker, isolating intact organelles from the synaptosomes and separating them on a linear density gradient. Using western blot, electron microscopy and mass spectrometry techniques I have identified the fractions which contain BEs.

Phd Candidate
Local synthesis of proteins is an important mechanism underlying the synaptic changes implicated in learning and memory but the regulation of the mRNA transport to those sites is not well understood. Based on the results of a proteomics analysis of RNA granules I used members of the DEAD box family of RNA helicases to characterize multiple types of RNA:protein complexes that transport and store mRNA in dendrites. I then showed that distinct types of RNA granules transport different mRNAs and are regulated through distinct signalling pathways, thus leading to specific responses of the cell to various stimuli.
