Investigated the role of Phosphatase and Tensin-like homolog (PTEN) protein in the regulation of ENaC channel activity using patch clamp. Single channel recordings indicate carbonylation of PTEN by a reactive aldehyde results in increased open probability of ENaC.

Developed confocal technique for visualization of ENaC channels located in the apical plasma membrane of renal epithelial cells. Initiated a lentiviral program utilized by three distinct laboratories. Created stable cell lines lacking period 1, endothelin-1 or gapdh. Knockdown of endothelin-1 resulted in a moderate reduction of ENaC alpha gene expression, but an increase in ENaC alpha protein consistent with inhibitory role of endothelin-1 on ENaC channel activity.Characterized glucocorticoid regulated endothelin-1 expression in renal collecting duct cells. Discovered that the hormone response element in the endothelin-1 gene contained a single noncanonical hexameric binding site responsible for high affinity MR and GR binding. Established library of proteins bound to the hormone response element or an E-box motif in the endothelin-1 promoter. Results show that period 1 binds directly to the endothelin-1 promoter E-box and negatively regulates its expression consistent with a stimulatory effect of period 1 on ENaC activity.Directly supervised honors research students and two graduate students whose projects were all developed as a direct result of dissertation work. Effectively negotiated discounts on major equipment purchases saving the laboratory thousands of dollars.
