The discovery of familial interferonopathy patients with mutations in the Perk gene has highlighted the importance of stress response pathways in the autoimmune process. PERK is a sensor involved in the integrated stress response (ISR) and the unfolded protein response (UPR). Two stress response systems that facilitate the restoration of homeostasis and cell survival.My PhDproject aims to investigate the effects of PERK point mutations on UPR and ISR signaling pathways, as well as to elucidate the role of PERK in interferonopathies, particularly in patients with systemic lupus erythematosus (SLE) and STING-associated vasculopathy with onset in infancy (SAVI). To do this, I use complementary methods such as flow cytometry, confocal microscopy and cell biology methods.Beyond these pathologies, I am interested in the impact of stress pathway regulation on cancer development, and how these signaling pathways might modify disease progression.
