I worked within a lipidomic research group at the University of Manchester. I developed a mass spectrometry assay capable of quantifying skin ceramides from tape strips. I applied this assay to discern the seasonal variation of ceramides in normal and acne skin, and to test the efficacy of various topical treatments aimed at restoring ceramide levels. The project required me to contribute to papers as well as writing Methods, COSHH and risk assessments for internal use. From this project I have UPLC-MS/MS method development experience. The role included significant amounts of liquid chromatography and mass spectrometry problem solving. I passed this knowledge on via lab presentations or by training other members of staff to use this assay. I also carried out maintenance of the liquid chromatography systems and mass spectrometers as well as managed the laboratory consumables.

Techniques used in this project include Maldi Tof CID Protein mass spectrometry, protein purification, bioinformatics and 2d gel electrophoresis and isoelectric focusing. External ProjectConducted offline Reverse Phase - MALDI ToF/ToF mass spectrometry of insect hormones coupled with bioassay to identify hormone effects on insect mating behaviour. This included extensive data analysis and interpretation with regard to experiments including Mascot searching, bioinformatics and calculation of false positive rates. Other techniques included insect dissection, protein extraction techniques.

I worked in a contract research organisation that carried out phase 1 and 2 clinical trials, specialising in treatments for respiratory and inflammatory diseases (specifically asthma and chronic obstructive pulmonary disease). My role included the following:- Assessing the severity of disease in patients using clinical methods such as: Spiriometry,Exhaled Nitric Oxide, Reversibility and Skin Prick testing for allergy. I explained the varioustesting methods and results to patients.- Processing both biopsy and sputum samples that could later be used for ELISA, Immunohistochemistry and RNA analysis.- Conducting differential cell counts on immune cells as part of patient screening for inclusion into clinical trials.- General Laboratory duties (sample storage, reagent preparation, electronic databasing of results etc).- Strict adherence to standard operating procedures.-All work was carried out to good clinical practice and good laboratory practice regulations.
