Phd Student In Breast Cancer Glycobiology And Therapeutics
My project involved exploring the impact of glycosylation on breast cancer progression. Specifically, understanding the role of high-mannose N-glycans in breast cancer and their potential implications for target therapies. During my PhD, I have gained exposure to numerous molecular biology techniques ranging from protein engineering to in vitro antibody effector function assays.

My roles mainly revolved around engineering and analysing antibody constructs.These included: PCR and molecular cloning, mammalian protein expression and purification, and glycan analysis by UPLC/MS. Other roles included mammalian cell line maintenance and inventory maintenance.

During this period I constructed gus knockout mutants of E. coli BAP1 through Lambda Red recombination and PCR targeting. This will assist future research in deciphering the structure of the E. coli glucuronide transport system proteins.Working at the National Key Laboratory enabled me to further develop my laboratory skills in bacterial genetic manipulation. More specifically I developed my laboratory techniques in: - Overlap extension PCR (including troubleshooting reactions)- E. coli transformation (via heat shock and electroporation)- Preparation of competent cells- Gel extraction and DNA purification- Bacterial cell culture- Enzymatic assayThe generation of this mutant strain will enable other researchers at Jiao Tong University and Bournemouth University to accurately measure changes in the functionality of the glucuronide transport system proteins, following directed genetic manipulations. Therefore, this research is extremely valuable to researchers in deciphering the structure of the E. coli glucuronide transport system proteins.To fund this research I was awarded two academic scholarships, including the highly competitive British Council Generation UK-China scholarship. My time working at the National Key Laboratory enabled me to thrive within a multicultural environment and allowed me to learn from experts in the field of molecular and cell biology. I developed my cultural adaptability, global communication skills, scientific understandings, resilience and confidence. Further, I set aside time to shadow more experienced researchers. From this, I became familiar with new skills including:- Western blotting- Protein purification - Cryo-electron microscopy
Undergraduate Dissertation Laboratory Student
During my undergraduate dissertation I employed genetic manipulation techniques and molecular cloning to assess the locations of molecular recognition within the E. coli glucuronide transporter protein. Sandwich Placement Bioinformatician
Throughout my placement I evaluated the performance of several taxonomic classification tools using a mock metagenome generated in-silico. From these analyses, I developed several classification pipelines which optimised tools for data processing, assembly, annotation and classification. These pipelines were applied to unknown arthropod metagenomes to identify the presence of pathogens and parasites which Dikerogammarus villosus and Dikerogammarus haemobaphes, may host. These pipelines were also documented and taught to other researchers for future use.
