During this period I constructed gus knockout mutants of E. coli BAP1 through Lambda Red recombination and PCR targeting. This will assist future research in deciphering the structure of the E. coli glucuronide transport system proteins.Working at the National Key Laboratory enabled me to further develop my laboratory skills in bacterial genetic manipulation. More specifically I developed my laboratory techniques in: - Overlap extension PCR (including troubleshooting reactions)- E. coli transformation (via heat shock and electroporation)- Preparation of competent cells- Gel extraction and DNA purification- Bacterial cell culture- Enzymatic assayThe generation of this mutant strain will enable other researchers at Jiao Tong University and Bournemouth University to accurately measure changes in the functionality of the glucuronide transport system proteins, following directed genetic manipulations. Therefore, this research is extremely valuable to researchers in deciphering the structure of the E. coli glucuronide transport system proteins.To fund this research I was awarded two academic scholarships, including the highly competitive British Council Generation UK-China scholarship. My time working at the National Key Laboratory enabled me to thrive within a multicultural environment and allowed me to learn from experts in the field of molecular and cell biology. I developed my cultural adaptability, global communication skills, scientific understandings, resilience and confidence. Further, I set aside time to shadow more experienced researchers. From this, I became familiar with new skills including:- Western blotting- Protein purification - Cryo-electron microscopy
