DePinho Laboratory, Medical Oncology, Dana-Farber Cancer Institute• Contributed technical expertise and project organization leading to successful completion of several concurrent studies on animal models of pancreatic and gastrointestinal cancer - Examined the combinatorial roles of four genetic lesions in the development of pancreatic ductal adenocarcinoma: oncogenic Kras activation and loss of three tumor suppressors p16(Ink4a), p19(ARF) and p53 - Investigated the role of LKB1 loss in gastrointestinal polyps in a mouse model and its impact on mTOR signaling - Elucidated the requirements for oncogenic Kras activation and loss of tumor suppressors p16(Ink4a) and p19 (ARF) in the initiation and maintenance of pancreatic cancer in transgenic mouse models • Established and independently managed multiple transgenic mouse cohorts, planned and carried out complex transgenic line crosses, genotyped and identified experimental and control groups • Harvested normal and tumor samples for parallel downstream analyses such as: gene expression, array comparative genomic hybridization, western blotting and histological processing, collected blood plasma for biomarker discovery • Optimized current procedures and created new protocols for immunohistochemistry following antigen retrieval in formalin-fixed and frozen tissues
