About Mahadesh Prasad A.J, Phd

I am committed, reliable, a biomedical scientist with excellent hands-on experience in vaccine development, molecular immunology, and Hematopoietic stem cell biology. I am a driven scientist, motivated to develop creative research designs and protocols that provide answers to complex biological challenges. My area of research is mainly focused on developing the vaccines for SARS-CoV-2 and ZIKA virus using Yellow fever-17D backbone. Additionally, I am extensively involved in understanding the molecular events involved in viral associated B- cell transformation and elucidating the role of early transcription factors in B- cell maturation and in formation of B- cell malignancies during early hematopoietic stem cell maturation using cell culture and mouse model. I am a confident, excellent project manager playing an important role in several independent projects resulting in publication in high-impact journals such as Nature, Blood, Journal of Virology, PLOS Pathogen and Oncotarget, etc. In addition to managing projects, I am continuously involved in grant application writing, manuscript writing, reviewing and supervising Ph.D. students, project students, and technical staff.     My postdoctoral research was mainly focused on elucidating molecular mechanism involved in B- cell transformation during Epstein Barr- Virus (EBV) infection.• Designed a novel project to explore the importance of EBV onco protein EBV nuclear antigen 3C (EBNA3C) in the formation of B- cell malignancies via inducing DNA damage response. • Performed EBV virus preparation and infection studies. • Optimized the study protocol to infect human peripheral blood mononuclear cells with EBV virus.• Generated number of different mutations in full length EBNA3C oncoprotein to identify the importance of different regions in B- cell transformation. • Performed flow analysis and proteomics studies on mammalian cells to understand transformation process during EBV infection. • Supervised project students for master thesis. • Reviewed manuscripts from major science journals. • Published research articles in high impact journals

    After submitting my PhD thesis to University of Mysore in 2009 June, I was invited as a visiting researcher to investigate the role of clinically important serum enzymes in development of cardiovascular diseases in domestic animals.More often domestic animals suffer heart related disorders which leads to prolonged illness and may even death. My project was mainly focused on evaluation of level of antioxidant enzymes particularly, Paraoxonase1 (PON1) enzyme in Canine suffering from Dilated Cardiomyopathy (DCM). PON1 is high-density lipoprotein associated esterase capable of hydrolyzing various organophosphates and protects low-density lipoprotein peroxidation. PON1 is essentially involved in prevention of cardiovascular diseases and the level is significantly decreased in myocardial infarction, liver disease and in oxidative stress. Our results form evaluation of serum samples from DCM with a varied illness showed that PON1 activity was significantly decreased in stage IV DCM, compared to healthy canine and stage II and III.

    Completed PhD in Biochemistry under supervision of Prof. Cletus JM D' Souza and Prof. Kemparaju.K, thesis entitled Substrate specificity of Paraoxonase1 (PON1) and their implication in cardiovascular diseases My thesis work represents the rapid method of purification of PON1 enzyme by advanced methods. My thesis work was mainly focused on identifying and synthesising novel potential atherogenic molecules such as fatty acid esters and cholesterol esters. These synthesised substrates were screened for substrate specificity of PON1. We extensively screened natural substrates such as Penicillin, Vitamin A as a substrate for Paraoxonase. During my PhD work we had collaboration with number of software firms to obtain clinical samples from their employees. We assessed the clinical samples form south Indians to monitor the susceptibility towards cardiovascular diseases
