• Major responsibility in cloning strategy development for different monoclonal antibody (Heavy chain and light chain) generation with different expression vectors.

• Worked on Antibody V-Gene Rescue Projects: Recovery VH and VL sequences from hybridoma cells.

• Knowledge of Western Blotting Using iBlot® 7-Minute Blotting System.

• Cloning strategy designing and construct development : Ligation dependent (Using restriction enzymes) and Ligation Independent cloning (Infusion cloning), Topo cloning

• RNA isolation and cDNA synthesis.

• Working experience on gene modification with Site Directed Mutagenesis.

• Primer designing, PCR, restriction digestion, ligation, transformation, Colony PCR.

• Genomic DNA and plasmid isolation and scale up of plasmids (Mini/Maxi/Mega/Giga prep) depending on the requirements for cell culture lab for cell line (Transient/Stable) development (Mammalian, insect cell lines)

• Linearization of circular plasmids for transfection for stable cell lines development and protein expression studies.

• Analysis of modified recombinant plasmids using sequencing and Agarose gel electrophoresis, Spectroscopy (A260/A280, A260/230 ratio)

• Performing nucleotide/amino acid sequence editing activities using different softwares like Vector NTI, Sequencher, Oligocalc, Primer3 Input, Signal P4.1 etc.

• Independent working and troubleshooting ability, worked on multiple projects in parallel.

• Successfully developed new research strategies applying self knowledge in current organization.

• Knowledge of NCBI Database analysis including BLAST, FASTA format, CDS, Database search, Allergen database of protein, Codon optimization.
