About Manon Chasles
Listed skills include Reproductive Biology, Molecular Biology, Cell Biology, Cell Culture, and 2 others.

Post Doctoral Researcher
Completion of a 2-year and 8-month contract as a post-doctoral researcher on a project aimed at analyzing the neuroendocrine, metabolic, and behavioral consequences of prenatal androgenization in sheep as part of a study of polycystic ovary syndrome (PCOS). This project was a collaboration between the Neuroendocrine Brain Development and Plasticity team at INSERM in Lille (59) and the MiDyNett team at INRAE in Tours (37).I developed and coordinated the various experiments, I also wrote the regulatory documents necessary for the experimentation authorizations (APAFIS referrals).I have set up a set of behavioral tests to characterize the consequences of prenatal androgenization among these, "novelty" tests (Open-Field, Novel Object), others to assess the cognitive skills (Detour Task), or a test to assess “optimism” (Judgment Bias Test).I was also able to analyze hormonal profiles, but also MRI images of brains as well as sheep ovaries (analysis via ITK-Snap software). Finally, I took samples from different animal tissues (fresh / fixed) and performed their immunohistochemical analysis using the QuPath software after image acquisition by a slide scanner.During this contract, I also participated in the promotion of my work through oral presentations or displayed at various scientific congresses (SNE, NeuroFrance, ..).My experience in animal experimentation, in particular with small ruminants, was able to be valued by participating as a speaker in two training courses promoting good practices in animal experimentation. During those trainings, I was able to explain how and why to seek the cooperation of the animal during the experiments, in particular the experiments including an evaluation of the behavior of the animal.
Phd In Biological Sciences - Speciality In Animal Reproduction
Management of a 3-years thesis project within the team « Neuroendocrinology of Sexual Interactions and Behaviors » at INRAE in Tours (France). This project aimed to characterize the consequences of prepubertal exposure to bucks on the reproductive function of the doe.I was required to develop and organize the various experiments on animals in coordination with the shepherds of the experimental unit as well as to draft all the regulatory documents required for experimentation authorizations (APAFIS referrals).I carried out blood samples, behavioral observations, participation in farrowing, analyzes of hormonal profiles (including use of the DynPeak algorithm), participation in radioimmunoassays of luteinizing hormone, samples and brain fixations as well as their analysis in immunohistochemistry, confocal microscopy.During this thesis, I was also involved in an international laboratory project associated with LIA-CABRAA (Goats as a model in Reproductive Biology and Animal Agroecology) in partnership with a Mexican research team located in Torrèon (Coahuila , Mexico). This team hosted me for a month in their research center, CIRCA (Centro de Investigación en Reproducción Caprina) in order to carry out experiments in relation to my research subject.During these 3 years, I also supervised many interns of different levels and coming from different universities. I supervised them in the field and I also supervised the writing of their theses and the preparation of their defense.
Internship (M2) - Analysis Of Genes Related To Lipid Metabolism In Bovine Intrafollicular Cells
This internship took place during my formation, Master's Degree in "Reproductive Biology" from the University of Tours.I was analyzing the expression of a family of genes called the tribbles in the bovine cumulus and granulosa cells.Techniques used: ovarian punction, in vitro maturation, cell culture, ADN extraction, RT-qPCR, immunofluorescence, confocal microscopy
