Post-Doctoral FellowUniversity Of Washington Nov 2008 - Dec 2013As a graduate student, I characterized the function of a novel DNA damage signaling protein, ultimately placing it in the 53BP1/SMC1 axis. As a postdoctoral fellow in the Loeb laboratory, I have explored the contributions of replicative DNA polymerases to mutagenesis, contributed to the development and evaluation of a platform for directed molecular evolution, and initiated an ongoing project to identify compounds to kill cancer cells by modulating the mutation rate. Over the past year, I have been developing Duplex Sequencing, a highly accurate next-gen sequencing technology that was conceived in the Loeb lab, for use on the following projects: 1) Probing DNA derived from primary tumors for rare subclones that could be sentinels of relapse; 2)Obtaining reliable sequence from FFPE archival samples; 3)Assessing very short-term mutation accrual as a consequence of environmental exposure.
