I lead the Protein Engineering team to continually improve the performance and quality of our polymerase, the key enzyme that makes Sequencing by Expansion work. The Seattle team is heavily focused on realizing the initial goal of Stratos Genomics: producing a robust chemistry for generating rapid, inexpensive sequencing that is the world standard for clinical diagnostics.

I lead a growing team performing R&D in support of Stratos Genomics' goal of creating a robust, reliable, and affordable platform for massively parallel nanopore DNA sequencing. My primary responsibilities are managing the work and direction of the group, communicating progress to our CEO, communicating changing company priorities back to the group, and coordinating efforts between Protein Engineering and the other development groups at Stratos Genomics.

I led a small team in efforts to improve our protein purification methods and to design new polymerase mutants, both in-house and in collaboration with our strategic partners.

Development of Duplex DNA Sequencing technology for applications such as subclonal detection of cancer-predisposing mutations, and highly accurate sequencing of formalin-fixed archival samples.

As a graduate student, I characterized the function of a novel DNA damage signaling protein, ultimately placing it in the 53BP1/SMC1 axis. As a postdoctoral fellow in the Loeb laboratory, I have explored the contributions of replicative DNA polymerases to mutagenesis, contributed to the development and evaluation of a platform for directed molecular evolution, and initiated an ongoing project to identify compounds to kill cancer cells by modulating the mutation rate. Over the past year, I have been developing Duplex Sequencing, a highly accurate next-gen sequencing technology that was conceived in the Loeb lab, for use on the following projects: 1) Probing DNA derived from primary tumors for rare subclones that could be sentinels of relapse; 2)Obtaining reliable sequence from FFPE archival samples; 3)Assessing very short-term mutation accrual as a consequence of environmental exposure.
