Profile bio

About Marcelina Klajner

I’m a researcher with 10 years of experience in life sciences like biophysics, molecular biology, and biotechnology. I’ve worked on few of the most challenging scientific topics at the top rated world’s universities: OSU (Ohio State University, USA) and UdS (University of Strasbourg, France).Until recently I led a part of a project at PAN (Polish Academy of Sciences, Poland) focusing on introducing biophysical methods like AFM and Fluorescence Polarization to examine the DNA-protein interaction. Before joining PAN I worked on cancer research (OSU, UdS) and was lucky to use the most innovative methods like FRET, Confocal Microscope or single molecule Optical Tweezers to observe the DNA damage and DNA repair processes. The first led to developing the new generation of anti‑cancer drugs, whereas the latter to understanding of self-repair mechanism preventing the cancer creation. My short adventure with Roche, where I worked before switching to science, allowed me to spend a great time with other engineers learning about the IVD analyzers. Successes included fast and effective fixing the analyzers in the hospitals where their reliable work is indispensable. Despite it being a fantastic job I quit it to evolve in new fascinating direction. Thanks to that I feel equally confident in science as well as in industry. However, I think that the perfect match is the connection of both.I’m truly passionate about my work and always eager to connect with other scientists or engineers. Colleagues know me as a highly collaborative person who can always be trusted.I can (and often do) work well alone, but I’m at my best cooperating with others.I’m always interested in hearing from former colleagues, managers, or just interesting creative folks, so feel free to contact me if you’d like to connect.

Listed skills include Molecular Cloning, Protein Purification, Afm, Fret, and 15 others.

Technical Research Specialist In Biotechnology And Molecular Biology

Institute Of Immunology And Experimental Therapy, Polish Academy Of Sciences

Wroclaw, Lower Silesian District, Poland

Key responsibilities:★Molecular Biology, Biotechnology & Biophysics• Molecular cloning• EMSA• ATP-ase assay• Unwinding assay• Cell culture maintenance• Protein purification• DNA-protein interaction• AFM• Fluorescence AnisotropyProject: Initiation of DNA replication in helicobacter pylori I worked on assembly of the H. pylori initiation complex, which is mainly based on oriC-DnaA interactions. H. pylori initiation complex is distinct from E. coli because of bipartite nature of H. pylori oriCs. The goal was to answer the question why some bacteria have bipartite origins and how the initiation complex is formed on such origins. Among others I applied AFM and Fluorescence Anisotropy to study how DnaA interacts with DNA.

Mar 2015 - Mar 2017

Postdoctoral Researcher

Columbus, Ohio Area

Key responsibilities:★ Biochemistry, Molecular Biology & Biophysics• DNA repair• Homologous recombination• Strand exchange assay• Molecular cloning• Cell culture maintenance• Nucleosome reconstitution• Protein purification• FPLC• HPLC• FRETProject: Determining RAD51-mediated Homology Search Mechanism in ChromatinMy work focused on physical aspects of DNA repair processes. It was the goal of my post-doc tounderstand the detailed mechanism of the human RAD51 protein homology search on nucleosome DNAduring strand exchange reaction. We have found that RAD51 remodels nucleosomes by unwrapping theDNA from the histone core in an ATP hydrolysis dependent manner.

Sep 2011 - Jul 2013

Phd Student

Strasbourg Area, France

Key responsibilities:★ Biophysics• DNA damage• Anti-cancer drugs• Single molecule• DNA manipulation• FRET• Optical Tweezers• Confocal MicroscopyProject: Organometallic Compounds as Anti-cancer Agents: Interaction with DNA and Migration in CellsMy PhD studies concerned three of ruthenium derived compounds (RDCs) which possess an anti-cancer activity. I showed that the complexation of DNA with RDC is reversible, measured the affinity constant of RDC for DNA and identified the conformational changes of DNA structure induced by the absorption of RDC using FRET system and single molecule Optical Tweezers. In addition I carried out experiments at the single cell level to understand the uptake of these compounds and their internal migration with confocal microscopy.

Oct 2007 - Jan 2011

Technical Service Engineer

Warsaw, Masovian District, Poland

Key responsibilities:• Medical device repair• Medical instruments installation• Technical support• Technical training• Medical DiagnosticsI was responsible for installation, maintaining, and repairing the IVD analyzers of Roche company. Additionally, I prepared technical training and provided the customers with support. I was working both in a team and individually.
