My activities were focused on the execution and implementation of the major biochemical assays used to measure the activity of the compounds on mitochondria isolated from murine organs, using an automated liquid handling platform (Tecan, Freedom EVO series). I also worked work in vivo with mice using the "Langendorff" model for measuring ex vivo levels of LDH released from heart after ischemia.Techniques used: excision of murine organs; protocols for isolation of mitochondria from different murine and human tissues ; measurement of mitochondrial respiration with oxygraph; biochemical assays (absorbance and fluorescence) on isolated mitochondria (swelling and mitochondrial calcium uptake); set-up of the same assays in 96wells format with robotic system; SDS-Page; Western Blot. I then worked on several research projects… Show more I took part, in collaboration with EIO Experimental Oncology, in a phase I-II clinical study with the histone deacetylase inhibitor valproic acid plus chemoimmunotherapy in patients affected by different types of tumor (mainly advanced melanoma). I was responsible of measuring experimentally the correlation between biological and therapeutic effect of the drug, using an innovative method to detect histone acetylation levels by flow cytometry. I then worked on several research projects focused on studying deregulation of chromatin modifiers in tumorigenesis. In particular, I investigated the role of Histone Deacetylases in tumor cell lines as potential target for new oncologic therapies, focusing on the screening of potential inhibitors (HDACi). Over the last two years I have been responsible for the development of an innovative technique based on the use of restriction enzymes in order to obtain a systematic mapping of nuclease accessible sites in the human genome.Techniques used: isolation of human peripheral blood lymphocytes; FACS staining of lymphocytes and cell lines (technique published); cell culture; drug treatment of cell lines; PCR; qPCR; cloning in plasmids; DNA and RNA preps from cells and tissues and analysis on agarose gel; SDS-Page; Western Blot; isolation and analysis of genomic DNA; DNaseI hypersensitive sites assays; Southern blot; clone screening and analysis of DNA sequences.
