My expertise, at the interface of chemistry and biology, was forged from 24 years as a researcher. I specialized in the production of high-quality macromolecules for structural biology experiments; specifically, purified proteins, peptides, DNA and small molecules. I also have expertise in the production of molecular probes, diagnostic tools that are useful for enzymology and high-throughput drug discovery. My research has been published numerous peer-reviewed journals based on the work I did in industry, graduate school and during a postdoctoral fellowship in France; truly an amazing experience.I am also passionate about science education. I believe that the next generation of scientists need excellent training, inspiration, and guidance and I have been fortunate to have had the opportunity to spend time teaching that generation in the classroom. I have also mentored a handful of students that have gone on to careers in biotechnology industries, endeavors that brought me a great sense of pride. Finally, I pleased to announce that I have started a new job at Tyra Biosciences. Tyra is a precision oncology company that is developing therapies to overcome tumor resistance and improve outcomes for patients with cancer. My past experiences in chemistry, crystallography, and protein biochemistry mesh well with the core competencies at Tyra, positioning me to make meaningful contributions toward achieving the company goals. It is also an excellent opportunity for personal growth and within the context of a supportive and amazing group of scientists.

Postdoctoral FellowCommunauté Université Grenoble Alpes May 2013 - Apr 2015Grenoble, FranceDNA damaging chemotherapeutics that are used in the treatment of cancers can become ineffective as DNA repair pathways are upregulated; therefore, compounds that exacerbate DNA damage are sought after as co-treatments with front-line chemotherapeutics. As a postdoctoral fellow I designed a molecular beacon that reports on non-enzymatic cleavage of DNA at abasic sites. The beacon is useful in high-throughput screening formats and I used it to identify several compounds from a chemical library that cause DNA cleavage at abasic sites. In addition, compounds that inhibit DNA repair enzymes are also sought after as co-treatments with front-line chemotherapeutics. To pursue this problem on both fronts, I established an international collaboration with Torrey Pines Institute for Molecular Studies to screen their mixture-based chemical libraries for compounds that inhibit the activity of a DNA repair enzyme, APE1.

Ph.D. StudentSan Diego State University Sep 2004 - Aug 2011San DiegoDissertation: Analysis of Holliday junction Binding CompoundsHolliday junctions (HJ) are formed during genetic recombination and are critical intermediates in DNA repair pathways. During my dissertation, I synthesized a family of analogous small molecules and demonstrated that they inhibit the resolution of HJs in DNA repair and DNA recombination assays. I demonstrated that the compounds had antibacterial activity and I used fluorescence-based quenching assays to model binding to the HJ (Mol. Divers. 2011 Rideout). In addition, I published work on a independent set of small molecules (Bioorg. Med. Chem. Lett. 2010 Ranjit) and a set of peptides (Peptides 2013 Rideout) that also inhibit HJ resolution. I also worked on a project to determine the structure of the inhibitors bound to a HJ. I pursued this goal by crystallography and NMR. As a result, I received training in the preparation of stable-isotope labeled peptides for NMR, basic 2D and 3D NMR experiments, and I strengthened my skills as a crystallographer. -

Classes were twice a week, three hours a day, and topics covered general biochemistry, enzyme kinetics, bioinformatics, protein purification, cloning and microbial genetics. In addition, I mentored four undergraduate researchers and one masters student who learned to perform assays in our laboratory using advanced techniques in molecular biology and biochemistry. -
Research Specialist IDecode Genetics Jan 2001 - Jan 2003Bainbridge Island, WaI collaborated on contract research with major clients as well and performed internally funded primary research, most notably on human tyrosyl-DNA phosphodiesterase 1 (hTdp1). I synthesized and characterized nucleotide and oligonucleotide fluorescent substrates for hTdp1. I developed protein purification methods, produced protein crystals, aided in refinement and solution of crystal structures involving hTDP1 which led to multiple publications (Nucleic Acids Res. 2004 Rideout; J. Mol. Biol. 2004 Raymond). In addition I managed supplies for the biochemistry department and maintained machinery (HPLC, FPLC, oligonucleotide synthesizer). -

Specifically, I carried out the synthesis, reverse phased purification and re-crystallization of complex. I also aided in complex characterization by 1H NMR and presented data annually at group meetings.
