Full-time cardiovascular and metabolic research in VLCAD deficiency (VLCAADD) for my master thesis. The present study sought to respond to the pressing need for a human cardiac model for the study of VLCADD and its underlying pathophysiological mechanisms. For that purpose, 2D human induced pluripotent stem cell-derived cardiomyocytes (hiPSC-CMs) models from control and VLCAD mutated patients were successfully generated and served as a physiologically relevant platform to reflect patient-specific therapy responsiveness. Morphological and functional characterization was conducted to validate the CM-model, investigate phenotypic differences and to assess incubation upon drugs.Laboratory techniques: proficiency in iPSC/iPSC-CM culture, Cytocypher contractility analysis,immunostaining, qPCR, Western blot.

The focus of this research was modelling the NPC1 disease in cerebellum organoids. As a part-time intern I applied genetically engineered tools, like CRISPR/Cas9 to generate a control isogenic cell line, correcting the I1061T mutation on the NPC1 gene of patient-derived iPSCs.Laboratory techniques: iPSC culture, lipofection, nucleofection, flow cytometry.
