Initially, I collaborated with Dr. Jacqueline Barton to study the iron-sulfur cluster-containing domain of primase, p58C. We found that this domain plays an unexpected role in coordinating primer synthesis, which ultimately informed a new model for polymerase communication at the DNA replication fork; these results were published in the journal Science. In addition to this work, I applied various biophysical and biochemical approaches to establish the role of domain rearrangements in primase catalysis.

As part of an NSF REU, I produced and purified mutagenized calprotectin and used isothermal titration calorimetry to determine zinc and manganese affinity to the transition metal-binding sites in calprotectin. Building on this biophysical characterization, I used S. areus growth assays to show that disrupting transition metal binding by calprotectin abrogated calprotectin's ability to inhibit bacterial growth.
