Gained experience in handling Schizosaccharomyces pombe yeast cells and Escherichia coli bacterial cells with expertise in their manipulation and cultivation. Got familiar with protein extraction and Western Blotting techniques. Conducted genomic DNA extraction, PCR, plasmid digestion, DNA cloning, and cell transformation in both E. Coli and S. Pombe for strain construction. Created a Schizosaccharomyces pombe cell cycle reporter strain using fluorescent tags on phase-specific proteins (G1, S, G2, and M phases). Additionally, generated reporter strains lacking sty1 and atf1 genes to explore their roles in cell cycle regulation. Skilled in operating a confocal microscope (Airyscan 2) and performing time-lapse experiments in reporter and mutant strains, followed by comprehensive data analysis of microscopy videos. Show less
