ATUM is a CRO that provides services in gene synthesis, protein engineering, protein technologies, and cell line development (CLD) supporting customers during basic research, drug discovery, preclinical and manufacturing, and regulatory. • Supervision of staff in four departments (Protein Expression, Purification, Analytical, Bacterial, and Yeast) - 15 total direct reports.• Assist customers in overcoming discovery and developability issues, collaborating with all groups within ATUM to achieve goals.• Working with the protein engineering group to provide material for projects for drug development.• Meeting external partners to further business opportunities.• As part of senior management, I met with owners to discuss the direction of the company and methods to improve the quality of services. • Provide staff with the opportunity to grow and develop as scientists and group leaders to manage projects through the facility.• Lead Technology development projects affecting all aspects of protein production including CLD.• Manage external partners, ensuring to meet the deadlines, delivering protein with the necessary quality and documentation for customers.

As a gene therapy company 4D Molecular Therapeutics produces a portfolio of AAV vectors specifically designed only to target specific cell types for gene delivery. • Supervision of staff (3 members of staff).• Reported progress of process optimization of several products to the executive committee.• Participated in discovery selection meetings for moving vectors forward reporting critical attributes that could affect developability.• Participated in IND enabling committee for an AAV vector to treat opthalmological diseases.• Managed and implemented the development of a modernized AAV upstream, downstream, and analytics for the production process to support internal and partnered programs.• Transferred new process to pilot plant and provided training to staff on each unit of operation.

Catalyst Biosciences harnesses protease technology to develop therapeutics in the areas of Hemostasis and the Complement pathway for treating bleeding disorders and age-related macular degeneration (AMD). • Supervision of Staff (3 to 8 members of staff).• Participated in program meetings chaired by the CSO.• Implemented the use of wave bioreactors at Catalyst Biosciences for the generation of biologics by transient transfection in mammalian cells, producing at least 240 liters of cell supernatant each week.• Expressed and purified, in high throughput format, proteases from E.coli for in vitro studies.• Established high-density cultivation of E.coli for the generation of developmental candidate material for in vivo studies.• Expression and purification of developmental candidates for in vivo studies and partnered programs.• Established HPLC Bioanalytics studying post-translation modifications, aggregation, purity and refold • Generation of a stable cell line for FIX production of final product and participation in tech transfer of project partnered with ISU.• Gained significant practical experience in protein purification using IEX, SEC, Affinity Chromatography, HIC, Reverse Phase, and AKTA purification systems, including software (Unicorn), whether purifying the target of interest from soluble or insoluble material for R&D, in vivo studies or IND enabling studies.
