Car Cell Therapy Research Assistant
Vancouver, British Columbia, Canada
Under the supervision of Dr. Megan Levings and Dr. Bruce VerchereDevelopment and validation of chimeric antigen receptor (CAR) regulatory immune cells for targeting stem cell-derived beta cells, with the aim of enhancing cell grafts for Type 1 Diabetes (T1D) therapy.Designed, executed, and validated a CRISPR Knock-in on a stem cell line. Confidently performed multi-colour flow cytometry (Fortessa, Symphony, Cytoflex) and Flowjo analysis, cell culture, ELISA, stem cell differentiation, and multiple cloning workflows (designing and building many CAR iterations). Other techniques and work involves:• RTqPCR• PBMC isolation • Immunomagnetic cell separation• Coaggregations of various cell types• Primary T cell differentiation and validation • Intracellular staining • STAT Phosphoflow• Legendplex (+ analysis)• Immunostaining and Confocal microscopy• FACS sorting • Cre-LoxP• Genotyping• Lentiviral Transduction• Lipofection• Designing and executing immunoassays• Data visualisation in PRISM/Graphpad• Culturing and preparation of cells for murine transplantations• GSIS• Preparation and proper handling of cytotoxic substances (STZ) for murine injections
