Conduct & interpret complex mass spectrometry characterization experiments to support biologics, immunology, neurology, dermatology & oncology discovery & development programs. Protein characterization of proteins, antigens, mAb, multi-specifics, peptides and novel protein formats using ESI mass spectrometers, performing intact MW, peptide mapping, PTM identification, phosphopeptide mapping, in-gel digestion, chemical conjugation incorporation. Arthritis biomarker quantitation, antigen epitope mapping, TMT sixplex labeling, various immunoprecipitation methods. Peptide mapping of antigens & antibodies, N & C-term analysis, PTM identification & specific location information including glycan occupancy using mass tags. Epitope mapping (epitope excision) to differentiate epitope binding sites of varying commercial & in-house Ab. Detection of IP’d MHC Class II peptide fragments from cell lysates. Detection of published and novel phosphorylation sites on a Protein Kinase from PMA & CD3/CD28 stimulated Jurkat cells. Performing and analyzing data from MASCOT searches. Established a disulfide bond peptide mapping method with optimized sample prep and mass spec conditions for two separate instruments to observe scrambled disulfides of mAbs, multi-specifics, antigens and novel protein formats. Developed a released glycan fluorescence mass spec UPLC/FLD/MS method to accurately assign structures and quantitate global glycan heterogeneity. Experienced in complex protein separations. Member of 1st responder team, responds to on-site medical emergencies. Also previously performed protein purification & refolding using affinity, IEX and SEC chromatographic methods from cell paste & suspension media. Very experienced with Thermo mass spectrometers including Orbi-trap Velos with ETD, Lumos tribrid with ETD Experience setting up third party customizations such as Newomics M3 emitter, Sonation column oven, New Objective emitter columns, Ion Opticks emitter columns and ESI Source Solutions. Experience with preparation for a new instrument installation and initial setup. Work in a team environment, including multi-functional and cross-site project teams.
