Project title: SREBP1 supports hormone-dependent cancers adaptation to endocrine therapiesSupervisor: Dr Luca MagnaniMy project focused on epigenetic-driven drug resistance in ERα positive breast cancer.Previous findings in the lab suggested that the transcription factor master regulator of cholesterol biosynthesis, SREBP1, plays a key role in epigenetic-driven resistance to aromatase inhibitor in ERα positive breast cancer.The project aimed at dissecting the molecular mechanisms of SREBP1 activation to validate it as driver of resistance, with the long-term goal to stratify patients to the most appropriate therapeutic option.

As Data Manager within the Oncology Clinical Trial Office (OCTO) at San Matteo Hospital - Pavia (Italy), my work referred to two main areas:1) Management of several procedures required for patients participation to multicentric clinical trials (Phase II and III):• patients’ visits scheduling and organizational planning, booking of instrumental exams, collection of medical reports checking their agreement with data quality standards • IWRS/IVRS calls to screen, randomize and assign drugs to patients • handling, processing, storage and delivery of biological samples to the designated laboratories • Serious Adverse Events (SAE) notification according to the designated procedure• drug storage and accountability2) Data Management:• prompt and accurate data entry in eCRF and query resolution according to GCP and SOP regulations• development of standard operation procedures (SOP) for internal use• delivery and report of patients' clinical exams to sponsors by designated electronic systems• creation of archives of data in paper and electronic format, checking their compliance with data management procedures

Project title: “Optimization of Alk-1 extracellular domain production and purification for structural and functional studies” Supervisor: Dr Claudia ScottiMy thesis project focused on the optimization of Alk-1 extracellular domain (Activin receptor-like kinase I) expression and purification, for structural studies and for future production of monoclonal antibodies for therapy.Indeed, Alk-1 is a TGF-β type I receptor, related to the genetic disorder Hereditary Haemorrhagic Telangiectasia type II as well as involved in many types of haematological and solid cancers. I cloned the fragment of interest and I expressed it in a bacterial host system (E. coli), testing several approaches and investigating both the soluble fraction and the insoluble inclusion bodies (both classical and non-classical inclusion bodies). After thorough troubleshooting, I successfully optimized the protocols: the high yield of protein obtained and subsequently purified by several liquid chromatography methods was suitable for crystallization studies. After the appropriate screening, initial optimization was performed to obtain crystals ready for data collection at the x-ray synchrotron.

MasettoThe group mainly aimed at elucidating the functional properties of inner hair cells in a mouse model and at investigating how ionic currents are involved in cellular communication. My specific work focused on learning the principles of the techniques used in the lab, shadowing the researchers in the day-to-day procedures and preparation of reagents for experimental set-up. Part-time experience, while attending lectures and sitting exams.
