Project title: “Optimization of Alk-1 extracellular domain production and purification for structural and functional studies” Supervisor: Dr Claudia ScottiMy thesis project focused on the optimization of Alk-1 extracellular domain (Activin receptor-like kinase I) expression and purification, for structural studies and for future production of monoclonal antibodies for therapy.Indeed, Alk-1 is a TGF-β type I receptor, related to the genetic disorder Hereditary Haemorrhagic Telangiectasia type II as well as involved in many types of haematological and solid cancers. I cloned the fragment of interest and I expressed it in a bacterial host system (E. coli), testing several approaches and investigating both the soluble fraction and the insoluble inclusion bodies (both classical and non-classical inclusion bodies). After thorough troubleshooting, I successfully optimized the protocols: the high yield of protein obtained and subsequently purified by several liquid chromatography methods was suitable for crystallization studies. After the appropriate screening, initial optimization was performed to obtain crystals ready for data collection at the x-ray synchrotron.
