    Postdoctoral scientist at the Biological Adaptation & Aging UMR 8256 (INSERM, CNRS, Sorbonne University). Aging is a progressive deterioration of cellular functions resulting in disease and death, but its molecular causes are not completely understood. Increased protein oxidative damage has been reported during aging in different organisms. The central hypothesis underlying my research states that protein damage is a major cause of cellular dysfunction during aging. Mechanistically, such damage would lead to critical alterations in the structure-function of key cellular proteins. In the recent years I have been involved in studies evidencing that the oxi-proteome (the sub-set of the cellular proteome targeted by carbonylation) during aging and age-related diseases is composed only by a restricted group of proteins, indicating that not all proteins have the same susceptibility for oxidation and propensity to accumulate at oxidized forms. Common processes such as increased oxidative stress, accumulation of protein damage and general metabolic dysfunction are suggested to be causally involved in aging. However, these processes have been mostly seen as independent events. A key question to approach is whether this deregulation is driven principally by oxidative damage of key enzymes. My research was aimed at defining the causal role of oxidized proteins in the human ageing process.

    As a graduate student (2006-2008) in Pr. Bernardino Ghetti´s lab (Indiana University, USA) my focus of research was the molecular characterization of a mutant ferritin that causes Hereditary Ferritinopathy, a neurodegenerative disease characterized by the presence of ferritin containing inclusion bodies and iron accumulation in the CNS. We found that the mutation induced conformational changes in the mutant ferritin leading to iron mismanagement and importantly iron-induced protein aggregation, consistent to what observed in patients carrying the mutation (J Biol Chem, 2008). Furthermore, we also solved its structure using X-ray crystallography, which support the structural basis of iron mismanagement observed (J. Biol. Chem, 2010). Importantly, we also identified oxidative post-translational modifications in the mutated protein and a free radical-mediated cleavage of the polypeptide chain, due to iron-catalyzed oxidation as a consequence of iron mishandling (FRB&M, 2012).
