The aim of the internship was to explore the diversity of a new class of beta-propeller enzymes. My duties consisted in cloning and expressing several enzymes found among different cyanobacterial genomes and evaluating their activity. I learned the basics of enzymatic activity assay and LC-MS/MS analysis.

I tackled the topic of enhancing heterologous protein production by fine-tuning the tRNA modificome. I learned basic RNA methods, such as total RNA isolation, acetylated and deacetylated tRNA isolation from yeast and urea-PAGE. Moreover, I generated in vitro translated mRNA reporter templates and performed translational analysis in cell-free systems

My main duties consisted of quality control assessment of the production process, chemical and microbiological analysis of raw materials and final products. Other than technical skills, I gained important soft skills such as communication, preparation and handling of official documentation, and decision-making.

The main objective was to characterize NLPPya cysteine mutants, a pore-forming protein present in plant pathogens. I gained proficiency in ÄKTA systems, in addition to which I did fluorometry assays, conductometry measurements, and performed protein-lipid interaction assays.

The aim of the internship was to obtain successful secretory expression of gluten protein in Pichia pastoris. The work included RNA isolation from rye grains, construction of gene casette via Gibson assembly, expression analysis, PCR, Gibson cloning, interpretation of sequencing data, tricin-PAGE, dot blot, affinity chromatography.
