This position was focused on my research project as a fourth year student. This project focused on validating a new twist to an existing (but old) way to measure reflected light and using it to track the Fv/Fm ratio over the growth curve of the centric diatom T. weissflogii & bacterium S. bacillaris. This project continued over my 4th year as a student as well (September 2019 - May 2020). This was my first major experience in independent microbiology, including the development of several new protocols.Experimental DesignThe Fv/Fm ratio is a calculation used to measure the ability of Photosystem II to absorb light in organisms. This project implemented a design to use an algicide to inhibit chl a. In doing so, we were testing to see if the much cheaper (albeit new) spectrophotometer with this method could achieve similar results to more accepted methods of phytoplankton spectrophotometry. Additionally, this ratio can be used to determine a relative `health` of the cell culture, depending on how strong it is. I coincided this protocol while also measuring cell count, absorbance, regular fluorescence, and Frrf data to track the measured Fv/Fm ratio over the expected growth curve of the organisms.
