    https://ncats.nih.gov/staff/hallmaI lead a team of research biologists that develop and optimize both biochemical and cell-based assays for automated small molecule high-throughout screening in collaboration with NIH intramural and extramural partners, including several NCI Chemical Biology Consortium (CBC) programs. My research portfolio has an emphasis on oncology, but covers a diverse range of disease other human pathologies and biology questions.My group’s in-house research programs include identifying therapeutic avenues for rare and drug-resistant cancers, adjuvant strategies for platinum-based chemotherapies, developing assays for understanding blood plasma drug metabolism, developing assays for in vivo and ex vivo small molecule target engagement, understanding the role of glutathione peroxidase in cancer, and the role of drug-blood-brain-barrier interactions that limit CNS penetrance.

    I have a diverse range of research interests at the NCI which have led to several intramural collaborations that aim to wed diverse discipline sets to tackle challenges in molecular pharmacology. I have three major areas of focus:Imaging transporter function at the blood-brain barrier. A collaboration with researchers in the National Institute of Mental Health has helped to develop a radiolabeled P-glycoprotein substrate to be used for PET imaging of blood-brain barrier (BBB) function. Recently reported bioluminescent imaging can be used to study ABCG2 function at the BBB.Collateral sensitivity of MDR cells. Identification and development of compounds that are selectively toxic towards multidrug resistant cancer cells. This project brings together a number of approaches, including mechanism of action, structure-activity analysis and I have also established a high-throughput screen of over 400,000 small molecules to identify new chemotypes. I was recently awarded a competitive NIH R03 grant to conduct this screen.Cisplatin resistance. Work continues on this important class of drugs. Designed whole-genome siRNA screen to identify genes associated with cisplatin resistance, identifying several new genes associated with cisplatin resistance/sensitivity.
