About Matthieu Gratia
I worked as post doctoral reseacher at the Curie institute in the biology / immunology after a thesis in virology.I wish to keep working in these domains and eventually enlarge my skills.My formation allowed me to specialize myself in biology and more recently in immunology. I did my thesis at the CNRS of Gif sur Yvette with the main objective to charaterized the implication of a viral protein in viral and cellular processes. Currently, I work to characterize the role of nuclear factor in the triggering of innate immune response in cells.My expertise lies in cellular and immunological process and I have a great interest in working in a scientific field that I can spread and shared.

Postdoctoral Researcher
Post-doctoral project with the collaboration between the teams of Dr.Mounira Amor-Guéret (UMR3348 Curie Orsay) and Dr.Nicolas Manel (U932 Curie Paris).More and more evidence links DNA damage, replication stress to immune responses. Moreover, scientists working in cancer field concentrate on the impact that can have immune responses in triggering of slowing down the establishment of cancers.My project is focused in the role a nuclear factor implicated in the replication progress and for which its deficiency can lead to cancers establishment. We have found evidences that this factor is implicated in the triggering of innate immune response in cells and want now to characterize what are the immune pathways implicated, and what are the causes of this response.
Phd : Initiation Of Translation And Rotavirus.
This is the continuation of my Master internship.Objectives :- Optimizing the quantitative system set during my internship.- Characterizing the effect of NSP3 during the translation of his own mRNA or of a reporter mRNA (luciferase system).- Characterization of rotaviral mRNA, and especially the untranslated region (5' and 3' UTR).- Attempt to purify recombinant NSP3 and use it for in vitro translation.Tools used :- In vitro synthesis and purification of mRNA.- Transfections into cells (overexpression of NSP3, electroporation of RNA reporters).- Virus infection (virus plaque assays).- Batch and FPLC protein purification.

The protein NSP3 of rotavirus is supposed to be involved into the translation of viral mRNAs and the shut down of the translation of cellular mRNAs. This model has been recently challenged.Objectives :Set up a quantitative system to study the rotaviral protein NSP3 dependent translationCharacterization of thermosensitive mutants on NSP3.Tools used :- In vitro RNA synthesis and purification.- RNA transfections in cells.- Quantification by luciferase assays / Western Blot.
