In short I produced and characterized actin from the Lokiarchaea using the Pichia pastoris protein expression system.In the lab of Dr. Ann-Christin Lindås (Wenner-Gren Institute, Stockholm University) the interest revolves around the life cycle of Archaea. In this project I produced and purified the protein: actin from the Lokiarchaea (an archaea that exhibits many eukaryotic traits). Genes were extracted from metagenomic sample and cloned into Pichia pastoris for either extracellular or intracellular protein expression. Induction times were established and proteins were purified using nickle affinity.

In the lab of Dr. Dennis Claessen (Associated Professor, Microbial Biotechnology & Health, Leiden University) I worked on major transitions in biology. We tried to create an artificial co-dependency between two separate cell lines. Valuable steps were made to achieve this goal. L-form (monomeric bacteria lacking a cell wall) were tagged with either eGFP or mCherry for easy tracking using confocal microscopy or flowcytometery. To the outside of the L-forms membrane peptides were attached to facilitate fusion between L-form bacteria. Co-dependency was achieved by knocking out a single gene that functions in both the tryptophan and histidine amino acid synthesizes routes, regaining of either the tryptophan or histidine synthesizing properties will be regained by substituting with E. coli genes for those amino acids.
