• Muscle endurance and atrophy model development in mouse and rat. Second author on a publication describing a novel method for the noninvasive imaging of muscle atrophy in a transgenic rat model. Expertise in the IVIS.
• Use of the Promethion continuous respirometry system and software to monitor and analyze metabolic rates of mice at rest, during exercise and in response to compound treatment. Completed a course at Sable Systems on.
• Muscle and vital organ dissection often in a large study team effort.
• Preparation and use of muscle lysates for analysis including western blots and glycogen content analysis.
• Histological preparation and sectioning of frozen skeletal muscle samples, followed byfluorescent microscopy.
• Mouse FDB Fiber preparation and infection with viral vector expressing GFP-tagged proteins. Experiments are done using automated fluorescent microscope taking time-lapse videos and observing GFP signal translocation.

Responsible for procurement and utilization of donor hearts (CTDN) and explanted diseased hearts from transplant recipients. Developing heart muscle slice technique for biochemical analysis, preparation of cardiomyocytes for culture and electrophysiological study.Point person in network of researchers with goal of most efficient use of this very valuable.

Contract position in which I was responsible for design implementation of in vivo experimentation. Skills used included small animal surgery, dosing, blood and bodily fluid collection, careful documentation and critical analysis of results.

• Performed CCI (Chronic Constriction Injury) Surgery producing mechanical allodynia. Implemented in vivo model of neuropathic pain using Von Frey hairs.
• Developed and validated rat Tail-Flick and Hargreaves models of nociceptive pain and tested in-house compounds for development.
• Prepared primary cell cultures of astrocytes and microglia from rat pup brains. Measured inflammatory cytokine liberation.
• Transfected SEAP reporter gene for colorimetric assay in TLR2 and TLR4 cells with generation of pure clones for plating and assay.
• Screened compounds in enzymatic plate assays: JNK2, 3 and PDE4, 10 were markers in for potential therapeutic area of neurodegenerative diseases.
• Used in-house lesioned animals or prepared rats in a commonly used model mimicking neural degeneration in the substantia nigra (i.e. Parkinson’s disease).

• Increased value of company by creating momentum within a stagnant program with crucial assay development. This fueled a SAR effort which led to an IND filing in Q3 2006.
• Developed ex vivo contractility models for the determination of smooth muscle motilin receptor agonist potency and tachyphylaxis.
• Measured the potency of three hundred motilin agonists.
• Developed a rabbit model of ex vivo tachyphylaxis following chronic in vivo exposure.
• Developed an Electric Field Stimulation model to study presynaptic motilin receptors.
• Performed gastric emptying measurements in dogs using a [13C] octanoic acid breath test.
