Specifically, I looked at the role of axonal and dendritic orientation relative to the direct current stimulus vector in neuromodulation. To examine the biophysical properties of these neuronal structures I used extracellular and whole cell patch-clamp recordings. In addition, I recorded evoked field potentials in neurons from post-surgical cortical slices from human brain tissue from patients suffering epilepsy using multi-electrode arrays. Thus, I could record from the different regions of the hippocampus while stimulating in one specific region tracking pathways of increased neuronal activity.

Summary: Electrophysiology, Patch-Clamp Recordings, Preclinical Testing in Diabetes Mouse Model, Molecular Biology, Immunohistochemistry, Potassium Ion Channels, Inner Ear ResearchResearch Description: In the Holt/Géléoc lab my research focused on identifying the biophysical properties of potassium ion channels in the inner ear. Using molecular biology I looked at developmental expression levels and potential effects of ion channel mutations on the function of the maturing hair cell. To examine the biophysical properties of these ion channels I use predominantly whole cell patch-clamp recordings.Research collaboration: preclinical research in pathophysiology and development of painful diabetic neuropathy. I conducted animal behavioral tests in leptin-deficient (ob/ob) mice which included intraperitoneal dosing of mice.In addition, I researched serotonergic and dopaminergic systems in Drosophila. Using immunohistochemistry I described serotonergic and dopaminergic morphology and used HPLC to screen for serotonin and dopamine levels. I screened for candidate serotonin receptors with PCR, HPLC, in situ hybridization and Western Blot. Further, in a collaborative study I used electron microscopy to locate serotonin receptors in brain slices of mice.
