Autism, depression, and anxiety disorders are associated with an imbalance in the serotonin system, which often involves changes in the function of the serotonin transporter (SERT). SERT interacts with and is regulated by integrins, transmembrane proteins that link the extracellular matrix to the intracellular cytoskeleton. My research focused on the physical and functional interactions between SERT and the integrin β3 subunit in the brain. Using mouse models that alter integrin β3 function, we 1) analyzed the signaling pathways involved in β3-mediated regulation of SERT activity and 2) evaluated the role of these signaling mechanisms in mouse behaviors linked to SERT function. These studies highlight the complex regulation of serotonin homeostasis and social behaviors by integrin β3 and reveal an important role for integrins in modulating risk for neuropsychiatric disorders. The orchestration of cell movements during embryonic development requires extracellular matrix (ECM) reorganization and the concerted action of planar cell polarity (PCP) proteins that regulate cell elongation and mediolateral alignment. These studies revealed that PCP-dependent integration of cell-matrix and cell-cell interactions function to coordinate the directed migration of polarized cell populations during development. Cadherins are a family of transmembrane proteins involved in cell-cell adhesion and play important roles in development, morphogenesis, and cancer. p120-catenin (p120) binds to the cytoplasmic tails of classical cadherins (E-cadherin, N-cadherin, etc.), which stabilizes cadherins and the entire adherens junction complex. My studies focused on the roles of different p120 isoforms in oncogenesis. Using shRNA-mediated knockdown of p120 in cell lines of various origin and tumorigenic potential, I… Show more Cadherins are a family of transmembrane proteins involved in cell-cell adhesion and play important roles in development, morphogenesis, and cancer. p120-catenin (p120) binds to the cytoplasmic tails of classical cadherins (E-cadherin, N-cadherin, etc.), which stabilizes cadherins and the entire adherens junction complex. My studies focused on the roles of different p120 isoforms in oncogenesis. Using shRNA-mediated knockdown of p120 in cell lines of various origin and tumorigenic potential, I identified a role for p120 in oncogenic transformation and dissected the suppressive and permissive roles of different p120 isoforms in tumorigenesis. This body of work underscores both the role of p120/cadherin complexes in cancer and the significance of p120 and cadherin isoform switching in oncogenesis. Show less
Postdoctoral Research Fellow In Cancer Biology
My research utilized 2D and 3D in vitro tumor models to analyze signaling mechanisms of oncogenesis, from which I was able to identify a role for p120-catenin in oncogenic transformation. During this time I wrote and was awarded an American Cancer Society Postdoctoral Grant, was an invited speaker at a Postdoctoral Research Symposium, and received several poster and oral presentation awards.
Postdoctoral Research Fellow In Cell Biology And Anatomy
During this short-term fellowship, I performed independent research on growth factor receptor signaling in breast cancer and assisted in manuscript preparation.
Embryology Laboratory Technician
As a laboratory intern, I was trained in human specimen analysis, tissue culture techniques, and instrument calibration. I also maintained laboratory supplies, prepared media, and performed quality control assessments of equipment and reagents.
