I am currently leading the Probe Development efforts which include creating affinity reagents for our proteomics platform.

My role as the Director of Technology Integration is to evaluate all aspects of our Nanopore Sequencing platform and help prioritize research activities to drive the platform’s performance to meet our product specifications. I help identify the key technical problems and help to organize and influence teams to focus on solving these issues. Taking the “30,000 ft” view of the system affords me the opportunity to recognize the gaps in our technology roadmap and present solutions to the project and management teams to maintain our product launch timelines.

I currently lead the Systems Integration Assay team which has several responsibilities, but at a high level we are responsible for pulling together all the various system improvements that the various functional teams are creating and make sure that they all work well with one another. In addition to the integration work, my team also has their own research projects that don't necessarily fall neatly into any of the functional team's areas of expertise.

I am currently leading the System Integration Assay group to improve the performance of our nanopore sequencing system. My responsibilities include identifying areas to improve our sequencing system and systematically altering the identified parameters to achieve improved accuracy and read length performance. This position includes managing a team of talented scientists and engineers, as well as working with other group leaders to improve parts of our system under their responsibility. This is a highly interdisciplinary position as I have to work closely with a wide variety of teams including engineering, software development, protein engineering and synthetic chemistry to achieve our target sequencing specifications.

My responsibilities as the Nanopore group leader included the production and assembly of our nanopore sequencing complex. I was also responsible for developing our next generation nanopore to improve nanopore sequencing performance. In addition to my biochemistry and biophysical characterization responsibilities, I was also responsible for evaluating the performance of these sequencing complexes on our sequencing prototype instruments. To accomplish these tasks I assembled a team of nine highly talented scientists with a wide variety of backgrounds and experience levels to meet the required performance specifications under very tight timelines.

I am in charge of expression, purification and characterization of target antigens for several of Medimmune's vaccine programs. I am using multiple expression platforms including E.coli, insect and mammalian cell culture for the production of these antigens, and characterizing the antigens with a wide variety of biophysical tools. I work closely with the vaccine teams to try and improve the immunogenicity of our antigens through a combination of approaches which include antigen modification and conjugation.• Led antigen production/characterization for three vaccine projects• Produced over a dozen antigens for vaccine trials • Identified antigen for high CD8+ T-cell response• Developed refolding protocol for difficult to produce antigen• Created new vaccine program to grow company pipeline

I studied membrane protein X-ray crystallography, where I uncovered a detergent dependent change of the subunit stoichiometry of the MscL channel. I learned various strategies for recombinant membrane protein expression, detergent extraction and protein purification. I was successful at growing membrane protein crystals using the standard sitting drop technique as well as growing crystals from bicelles. • Discovered detergents could reorganize the MscL stoichiometry from pentamers to tetramers• Introduced the yeast expression system (P.pas and S.cer) to the laboratory• Resident expert in static light scattering for protein mass analysis (SEC-MALS)

I studied epithelial ion transport of the Solute Carrier 26A (SLC26A) family of proteins using fluorescent microscopy as well as patch clamp analysis. The impact of disease causing mutations on ion transport was monitored, as well as how the WNK (with no lysine) kinases regulated ion transport in the epithelial membrane. • Identified SLC26A9 as a Cl- channel rather than transporter• Discovered cell surface expression of SLC26A9 is regulated by the With No Lysine Kinases (WNK)

As a graduate student I studied two human diseases, Cystic Fibrosis and Congenital Chloride Losing Diarrhea. I studied the molecular mechanism(s) of these diseases and found that the proteins responsible for these diseases physically associate and mutually activate one another. I studied the effect disease causing mutations in these two membrane proteins had on the folding and function of the transporters, and the impact these mutations had on epithelial ion transport.• Discovered disease causing mutations in SLC26A3 resulted in inactive protein due to protein misfolding• Identified a physical association between SLC26A3 and they Cystic Fibrosis Transmembrane Conductance Regulator (CFTR) which results in mutual activation of ion transport• Helped to elucidate the structure of CFTR Nucleotide Binding Domain 1 with and without ATP
