Experienced protein chemist and biophysicist with >10 years experience purifying, conjugating, and characterizing challenging tissue-derived/endogenous and recombinantly expressed protein targets to generate fit-for-purpose proteins which support programs and advance projects.Additional expertise in:Project management Molecular biology AKTA/Unicorn method development/optimization Bacterial and mammalian cell culture Biochemical & plate-based assay development and transfer New target evaluation Supervision and mentorship of junior scientistsI currently work in the pharmaceutical industry as a protein biochemist purifying and characterizing challenging proteins. My work at Prothena involved all aspects of drug discovery from target evaluation and discovery biology to mAb developability/stability studies and IND report writing. Key accomplishments and contributions: -Principal protein scientist on 3 campaigns, significant contributions to entire pipeline-Purified >100 proteins and mutants to support components of all aspects of drug discovery: target evaluation, discovery biology, characterization, development, and manufacturing-HCP assay (development, data collection, analysis, IND report)-in-vivo efficacy biochemical evaluation-in-vivo disease model biochemistry-in-vitro disease model biochemistry-Lot release assay (development, data collection, analysis, report writing, assay transferred to GxP)-mAb developability and stability study design, data collection, and analysis-Immunoassay development (IP, immunodepletion, etc)-In-house bioconjugation and bacterial expression expertMy post-doctoral work involved studying the structure and function of epigenetic readers and writers.Key accomplishments: 3 published manuscripts (see below), crystal tray setup and screening, protein modeling, restriction/gateway cloningMy graduate research focused on the aggregation of microtubule associated protein (MAP) Tau. Tau fibrils are a hallmark of 20+ neurodegenerative diseases. My doctoral work provided insights into conformational ensembles and conformational evolution through the use of a variety of techniques like TEM, FRET, fluorescence anisotropy, and cell-based assays to illustrate the need to further investigate how fibril strains and heterogeneity can lead to different phenotypic presentations and clinical consequences.Key accomplishments: 4 published papers, supervised >5 undergraduate thesis projects, purified >21 unique proteins (>50 including mutants)

ScientistRegeneron Dec 2021 - Jan 2023Tarrytown, New York, UsI lead the research reagent protein purification subgroup. My group is responsible for purification and characterization of high quality recombinant proteins which are used as research reagents and in the development of novel technology. We utilize a variety of biochemical techniques and chromatographic processes to enrich tagged and tag free protein preparations derived from conditioned media and cell pellets. These media and cells are derived from all commonly used expression systems including samples produced in bacteria, insect, human, and CHO systems. We are responsible for ensuring timely delivery and reporting on these molecules in order to support many different departments and request types. 📑Generated SOPs, workflows, and go/no-go decision trees for use by my junior direct reports✔️Set up the lab space for use by a new group; purchasing and installation of smaller equipment and instrumentation, outfitting benches, stations for specific processes, etc👩🔬👨🔬Train and mentor 1 direct report from novice to production quality 🧪Streamline existing departmental processes and protocols for efficient production of high quality research reagents -
ScientistProthena Corporation Plc Mar 2020 - Nov 2021Dublin, 2, IeAs a Scientist in the Protein Sciences group I was responsible for isolating and characterizing proteins from a wide variety of recombinant and tissue-derived sources. This role contributes to many neurodegenerative campaigns from target discovery and evaluation through development and manufacturing support by providing technical and conceptual biochemistry and biophysical expertise.• Develop purification and enrichment strategies to isolate proteins and antibodies from natural and recombinant sources• Provide material support and technical/conceptual expertise in the development of seeding models in-vitro and in-vivo across several programs• Provide conceptual input and technical biochemistry expertise to study protein aggregation and misfolding and characterize biochemical/biophysical properties of isolated proteins and protein aggregates• Contribute to supervision and mentorship of lab members within the Protein Sciences group• Interface with multiple discovery projects and matrix teams across the organization to identify and help evaluate potential new discovery targets• Identify and manage CROs to complement in house capabilities -
