About Marcus Gibson
I am chemical biologist with a broad range of expertise in end-to-end research and development of next-generation therapeutics. As a Principal Scientist in Computational Genomics and Data Sciences at Omega Therapeutics I am responsible for developing platform capabilities with respect to mRNA and protein design.Previously, as a formulation scientist at Omega, I developed the tools to deliver nucleic acid APIs for in vitro and in vivo studies, as well as the analytical methods to characterize delivery technologies from a QC standpoint as well as in-vivo distribution and activity. These have included DLS, FFF-MALS, flow cytometry, PCR, IHC, and fluorescence microscopy, amongst other techniques.I am a strong cross-functional team-member. I very much enjoy working with others and mentoring younger scientists, and my broad experience in chemistry, molecular biology, and bioinformatics allows me to work and communicate across scientific disciplines.

Natural products (molecular compounds made by living cells) make up a majority of the drugs that we use to treat various diseases from bacterial and viral infections to cancer. Many of these natural products require at least one precise chemical modification that makes them both potent drugs and also very difficult to synthesize in the lab. In the Seyedsayamdost Lab, I am studying the organisms that make these important compounds, the enzymes that are responsible for carrying out difficult chemical modifications, the genes that encode for and regulate their production, and the chemical details of how these molecules interact with their targets.In particular, I have worked on a team investigating the in vitro biosynthesis of glycopeptide antibiotics. These molecules include vancomycin, a drug of last resort and one of the most important antibiotics worldwide. Our team has recently demonstrated in vitro installation of the second bisaryl ether crosslink in the vancomycin biosynthetic pathway.

The Wood-Ljungdahl Pathway for acetogenesis is the most efficient carbon fixation pathway found in nature, utilizing a host of complex transition-metalloenzymes to reduce two molecules of carbon dioxide to one molecule of acetate. The principal focus of my research was a novel oxalate metabolizing enzyme, oxalate oxidoreductase (OOR), which enables its host organism, M. thermoacetica, to grow exclusively on oxalic acid via the W-L pathway.I determined multiple X-ray crystal structures of OOR, with and without substrate bound, that allowed me to gain insight into the unique oxalate chemistry of this enzyme. I was further able to carry out a bioinformatic analysis that allowed me to speculate with regard to related enzyme that are found in both similar and substantially different microbial environments.
