Now retired from academia and semi-retired as a consultant (It takes a lot to persuade me that today's not a good day to go walking, cycling, skiing, birdwatching etc) However I have extensive experience of the development of antibody based therapeutics including recombinant and bispecific antibodies. An inventor of the first fully humanised antibody CAMPATH (alemtuzumab) and several other humanised therapeutic antibodies to antigens such as CD3 (TRX4 otelixizumab) and CD4 (TRX1). Alemtuzumab is used off-label for organ and marrow transplantation and was approved for treatment of B-CLL from 2001. The antibody was later reformulated and repurposed under the tradename Lemtrada for use in the treatment of Multiple Sclerosis. I was also involved in some of the earliest clinical uses of CD3 as a monovalent and also as a bispecific (CD3xCD19) therapeutic agent.My subsequent work concentrated on antibodies with modified Fc properties and some of these modifications found their way into antibodies such as Tolerex / GSK's otelixizumab, and several of Pfizer's antibodies such as tanezumab, ponezumab, RN316 and RN564. The migraine treatment fremanezumab also includes one of these Fc modifications. In collaboration with the UK NHSBT we also investigated the use of a modified antibody to HPA-1a antigen for treatment of feto-maternal allo-immune thrombocytopenia (FMAITP).I've also worked as a consultant or on SABs on many commercial aspects of humanisation, deimmunisation, effector function optimisation and other aspects of antibody engineering for human therapy.I was more heavily involved including as an investor in the commercial early phase startups of Absolute Antibody Ltd and its spinout mAbSolve LtdIn academia I spent the years 1990 until my retirement in 2014 organising, lecturing and examining undergraduate courses in infectious diseases and immunology. In later years I also got involved in university politics serving as an elected member of Cambridge University's Council and also on its Finance Committee. Additionally I've experienced and influenced the evolution of Cambridge University's technology transfer policies over the years 1981-2014 (including UK Government's NRDC becoming the privatised BTG).Specialties: Humanisation and also Fc effector function modification.
