In 2001, I discovered the ancient enzymatic machinery that allows metazoan cells to monitor O2 abundance in their environment. The article was recognized as essential for the 2019 Nobel Prize in Medicine or Physiology. Together with the follow-up study published in 2002, my work with Dr. Kaelin also paved the way for the development of drugs that mimic the hypoxic response by inhibiting HIF prolyl hydroxylases. In 2007 my group reported for the first time microRNAs that are involved in the hypoxic response including mir210, the quintessential hypoxamir. In 2020 we described lincNORS / MIR193BHG, an RNA regulator of cellular sterol/steroid homeostasis. Remarkably, common genetic variations at this genomic location are associated with differences in sexual maturation, growth and cholesterol level. As of 2024, over 60 GWAS studies have “hit” the locus, an invitation from nature to look closer at this fascinating region.
