* Development of Adoptive T cell therapy for the treatment of refractory viral infections for transplant and immuno-compromised patients* Generation of virus specific T cells and Characterizing them using multi-color flow cytometry and RNA sequencing. * Development of Universal coronavirus specific T cells for adoptive T cell therapy.* Translation of multi-virus specific T cell therapy to clinic and submission of first in-house T cell clinical trial at Columbia University Medical Center.*Use of immunological techniques such as isolation of primary immune cell subsets, primary cell culture and expansion, transfection and viral transduction of immune cells, multicolor flow cytometry and cell-based functional assays for development and characterization of antigen-specific T cells.

1. Research project Design & Development: Designing and executing in-vitro experiments using breast cancer cell line and in-vivo experiments using project specific transgenic mouse models to study breast cancer biology. 2. Assay Development: Development and optimization of new in-vitro and in-vivo assay to test various hypothesis and establish standard protocols.3. Lab manager and lab safety officer: Maintain laboratory inventory, responsible for communication with sales representatives for pricing and order of reagents and communication with technical support staff for troubleshooting and lab instrument maintenance. Manage lab safety records, write SOP for bio-hazard and toxic reagents. 4. Grant and manuscript writing: Write different institutional research grants and manuscript related to project.5. Mentoring: Mentoring undergraduate research students which require designing project, teaching them new cell based in-vitro techniques and train them to present data at various domestic undergraduate research conferences.

Running cell and molecular biology lab: 1. Teaching theory and principle behind various cell and molecular biology experiments and techniques, plan experiment2. Plan experiments, prepare regents and treat cells prior to lab. Guide students to perform experiments and explain results of experiments.2. Teach students to design, write and conduct scientific projects by reading primary literature.3. Design and grade quiz, mid-term exam, group project proposal, presentation and lab reports. 4. As Senior research assistant, I was responsible for training and supervising the new employee and in charge for managing lab inventories and shipping the final product. Supervised and trained new employees to conduct Ion torrent next generation sequencing after completion of the aptamer selection process. I was also responsible for managing individual project from project design, scientific consulting with customer, target selection, aptamer selection and validation along with writing project report and shipping final product to the customer.

Development and optimization of Parallel SELEX to generate aptamersParallel Selex (Systematic Evolution of Ligands by EXponential enrichment) is a process of selecting specific oligonucleotides to the target protein. I have worked on the development and optimization of massive parallel SELEX using proprietary high-throughput aptamer selection protocol to yield high affinity aptamers for proteins, peptides, small molecules and whole cell. Developed methodologies to generate highly affinity RNA aptamers. During the project, I developed expertise on protein conjugation chemistry, aptamer characterization by measuring affinity (Kd value) using SPR. (Surface plasmon resonance) molecular cloning, and ion torrent next generation sequencing.

Project:Identification of microorganism and viruses by mass cataloging of RNAsAs an intern at BioTex Inc., I worked and optimised “universal” assay which can be used to identify bacteria, fungi and viruses in the human samples such as sputum, saliva and blood. Process involved amplification and in-vitro transcription of bacterial or viral Ridosomal RNA followed by MALDI-TOF mass spectroscopy. The project lead to successful collaboration with with Methodist Hospital at Texas Medical Center, Houston to identify infecting organism in the clinical samples collected from septic patients. The work resulted in a book chapter. Techniques used: PCR, Gel electrophoresis, PAGE, In vitro transcription and MALDI-TOF mass spectroscopy.
