Jan 2017 - Present

Molecular Biologist

University Of Ottawa

Ottawa, Canada Area

• Performed molecular cloning and developing transgenic construct to generate genetically modified flies.

Apr 2016 - Jan 2017

Research Associate

Ottawa, Ontario, Canada

•Used CRISPR/Cas9 genome editing techniques to modify the expression or sequence of key factors to study pathways.•Studied pathways of stemness using real-time PCR, western blot, luciferase assay on genetically manipulated cells and tissues.•Finded promoter region of genes contributing in Wnt pathway, finding cic-acting elements through bioinformatics studies. Jan 2014 - Jan 2017

Senior Postdoctoral Fellow

Ottawa, Canada Area

• Worked on stem cells, primary cultures and cell-lines• Examined the potential of dendrimers nanoparticles in cell studies• Produced and used letivirus and adenoassociated virus for in vivo and in vitro gene transfer• Applied small molecules and drugs in vitro to develop new therapeutic approaches• Performed studies on micro-RNA and profiled them using microrray• Performed histological studies• Trained 11 junior researchers and helped them with project design and troubleshooting and technical supportMain project 1: Investigating mechanisms underlying atrial fibrillation by modifying the expression of a key triggerAdenoaccociated virus is an efficient tool for in vivo gene transfers to study a key trigger of atrial fibrillation. However, lentiviral transduction is sufficient for in vitro gene transfers but it did not act as a competent vehicle for in vivo studies investigating the underlying mechanisms of atrial fibrillation. The promoter regions of genes involved in neural transmission are helpful tools in pinpointing the cells that play role in the disease. Main project 2: Comparison of miRNA content of cardiac stem cells in situ and ex vivo The miRNA content of a homogeneous population of cardiac stem cells varies depending on their source. Some of the factors that alter the miRNA profile of these cells include the age of the animal CSCs were obtained from, and their origin weather they have been isolated from tissue explants and ex vivo cultures or from fresh tissues, in situ. Inducing an miRNA expression pattern that enforces endothelial-mesenchymal transition could make these cells more beneficial in cell therapy approaches to prevent or reverse the cardiac remodeling following myocardial infarctions.

2015 - 2016 ~1 yr

Postdoctoral Fellow

Waterloo, On, Canada

• Analyzed promoter region of Glucocorticoid Response Element gene in Zebrafish• Identified regions of promoter that reveal environmental impact on zebrafish• Conducted reporter studies on zebrafish embryos and cell-lines, and purified various proteins• Trained 8 junior researchers in experimental design and trouble shooting experiments, and acted as a resource for their questionsMain project: Analysis of the promoter region of Glucocorticoid Response Element gene. Promoter region of Glucocorticoid Response Element gene consists of a 3.5 kb region including one glucocorticoid response element, one SF-1 site which is a key player in the development and differentiation of steroidogenic tissues, and an estrogen response element.Promoter region of steroidogenic acute regulatory protein (StAR) gene consists of a region of about 2 kb including two estrogen receptors, two SF-1 sites and a metal-responsive element binding site. These cis-acting elements are crucial in environmental impact on zebrafish. Both promoter regions are functional in zebrafish embryos and in vitro, acting as efficient regulatory elements. These genes are ubiquitously expressed during the early stages of embryogenesis especially within muscle and cardiac cells.
