To overcome this issue, I employed enzyme engineering strategies to effectively enhance the activity of a thermostable phytase. This research was part of the broader effort to explore innovative environmental applications for phytases, aiming to recycle phosphate from biobased products, slaughterhouse byproducts, and waste streams.- Performing rational design approaches on 3 microbial phytases (Debaryomyces castellii, rphyXT52 seq and Buttiauxella sp.) in order to improve their enzymatic performance (protein modelling & docking).- Examine how amino acids of the binding pocket contribute to the catalytic activity. - Constructing single-site saturation mutagenesis libraries of the 3 enzymes.-Phytase purification and characterization. -

In RAW264.7 cells, the expression of Smurf1 was monitored after MNV infection via Western-Blot and RT-PCR. Moreover, the interaction of Smurf1 with MNV was studied using Co-Immunoprecipitation technique. Immunofluorescence analysis enabled the monitoring of the stained ubiquitin protein levels after MNV infection in RAW264.7 cells.
