About Mohammed El Bachiri

I have a strong enthusiasm for leveraging protein engineering to forward innovation and solve complex biological challenges. My multidisciplinary scientific background in both experimental and computational biology allows me to design smart proteins for industrial applications.Proven track record in successfully translating computational designs into functional proteins and I'm committed to making a positive impact in developing innovative technologies through the power of computational biology and AI. Currently developing green technologies in enzyme engineering for Bioimpulse project (BIOIMPULSE) with Resicare, focused on innovating and creating new bio-sourced adhesive resins using next-generation biocatalysts.

Currently a member of the bioimpulse project working as an enzyme engineer. My role in the project is to engineer two enzymes that are of great interest in creating a new adhesive resin free of substances of very high concern (SVHC).Main tasks : - Use of modern computational methods (modelling, docking, molecular dynamics and protein design) for rational enzyme engineering.- Create rationally designed mutant libraries for optimising the protein fitness (enzyme activity) and to get a further understanding of sequence-function relationship.- Leveraging cutting-edge artificial intelligence (AI) methods (proteinMPNN, SaProt... etc) for enzyme stability design.- High-throughput screening of best active mutants.- Enzyme characterization.- Use mutant libraries data for machine learning guided enzyme engineering.

"Protein engineering of phytases for emerging environmental applications"Laboratory: ABBT Team of Prof. Dr. Ulrich Schwaneberg. In the course of my master's thesis project, I addressed a challenge prevalent in the feed industry, particularly the denaturation of phytase (supplemented in seed-based feed additives) during the pelleting process. To overcome this issue, I employed enzyme engineering strategies to effectively enhance the activity of a thermostable phytase. This research was part of the broader effort to explore innovative environmental applications for phytases, aiming to recycle phosphate from biobased products, slaughterhouse byproducts, and waste streams.- Performing rational design approaches on 3 microbial phytases (Debaryomyces castellii, rphyXT52 seq and Buttiauxella sp.) in order to improve their enzymatic performance (protein modelling & docking).- Examine how amino acids of the binding pocket contribute to the catalytic activity. - Constructing single-site saturation mutagenesis libraries of the 3 enzymes.-Phytase purification and characterization.

"Role of an E3 ubiquitin ligase in the autophagic response following infection by Norovirus"Laboratory: Center for infection and immunity of Lille.Evaluation of the role of Smurf1 (E3 ubiquitin ligaseà) during murine Norovirus (MNV) infection. In RAW264.7 cells, the expression of Smurf1 was monitored after MNV infection via Western-Blot and RT-PCR. Moreover, the interaction of Smurf1 with MNV was studied using Co-Immunoprecipitation technique. Immunofluorescence analysis enabled the monitoring of the stained ubiquitin protein levels after MNV infection in RAW264.7 cells.
