This project included understanding molecular biology of pathogenesis of Mycobacterium tuberculosis. Towards this, my aim was to generate successful knockouts of seven non essential Serine-threonine eukaryotic like protein kinases (STPKs) in virulent strain of Mycobacterium tuberculosis (Mtb). In my experimental work, I used the method of specialized transduction to generate knockouts of seven STPKs. This method of specialized transduction employs the use of conditionally replicating temperature sensitive phasmid replicons as delivery vectors to facilitate gene transformation at higher frequency. In this procedure a gene of interest, disrupted by a selectable marker, is cloned into a conditionally replicating (temperature-sensitive) shuttle phasmid to generate a specialized transducing mycobacteriophage. The temperature-sensitive mutations in the mycobacteriophage genome permit replication at the permissive temperature of 30 °C but prevent replication at the non-permissive temperature of 37 °C. Transduction at a non-permissive temperature results in highly efficient delivery of the recombination substrate to virtually all cells in the recipient population.

Title- Molecular Understanding of Brain : Analysis of Long Term Potentiation (LTP). During this project, focus was on two main parts: First, was to study the phylogenetic evolution of Long Term Potentiation- LTP is a neural mechanism of synaptic transmission that underlies the formation of memory and learning in an organism. Sequence alignment studies were done on chosen sequences of eleven molecules of LTP from some organisms and phylogenetic trees were constructed. Alignment scores were calculated for every single comparison and a table was formulated indicating the variation of a particular protein among the selected organisms.Second part was about the creation of a collective database of all the molecules of the brain - Molecular Database of Brain (MDB), that includes all the pathways of the brain along with details of every gene, protein and ligand participating in that particular pathway. MDB designed to support both experts and novices point of view consists of details of gene (location, Chromosome number, size and related database cross references); details of protein (symbol, PDB ID, function, size, molecular weight details, link to Uniprot, and to see the complete GO annotations) and ligand (Charge, Formula SMILE and InChikey notations).

Title- Protein Ligand Binding studies using different NMR experiments.The experiment’s main aim was to find out a particular concentration of BSA (bovine serum albumin), a protein and fluorinated tryptophan (5F-Trp), a ligand, at which they can show maximum affinity and thus can be useful in the drug delivery systems. To know the maximum affinity of protein ligand complex, various concentrations were tried and observed under NMR spectra. Moreover, Diffusion coefficient and Dissociation coefficient of BSA and 5F Tryptophan from variety of NMR experiments were calculated.

Title- Expression of mutant form of glycoprotein in Pichia pastoris and checking for its activity. The base of the experiment was to focus and exploit the role of a glycoprotein which is expressed in high amounts in immune cells and plays an active role in survival of the transplanted organs by inhibiting T-cell proliferation. However, it has been showed that activity of this protein is relied on the backbone of the protein, which is made of many long glycan chains. These huge glycan chains mask the epitopic region of protein and thus interfere in the suppression of Tcell proliferation. Thus my project was mainly concentrated to express the mutants form of this protein, both single and double mutant ( at two specific glycosylated sites) in Pichia pastoris and to check the activity of mutated protein, whether any mutant binds actively and suppress T-cell proliferation.

Title - Designing of Lipid microspheres and their use in drug delivery system The main aim of the project was to check the lipid microspheres as a potential of target drug delivery. Thus its viability and potentiality were checked to analyze the suitability of the drug carrier, for which I used lipid microspheres (stearic acid). Studied Potentiality of Lipid Microspheres as a protein delivery system. . The characterization of stearic acid microspheres was done by -Particle size analysis, Entrapment efficiency and In-vitro release.
