The purpose of the research was to investigate the role of the different gene classes and isoforms of starch synthases in starch biosynthesis. The long-term goal is to improve the quality and fermentive yield of starch in barley malt. The experimental work included:- Molecular cloning using codon-optimized genes for E. coli expression- Protein purification using Äkta FPLC - Protein crystallography- Enzyme kinetics using UV-vis spectroscopy, capillary electrophoresis, MALDI-TOF MS, UPLC, and NMR- Fluorophore labelling of glycans by reductive amination- Chemoezymatic synthesis of the substrate, ADP-Glucose - Agrobacterium tumefaciens mediated plant transformation of barley embryos with starch synthase mutantsI was during my employment introduced to the use of electronic laboratory notebooks (ConturELN)

The purpose of the research was to perform structure-function studies of retaining and inverting mannosyl transferases. In particular mannosylglycerate synthase from Rhodothermus marinus was used as a model system to study catalytic mechanisms of glycosyl transfer. The experimental work included:- Molecular cloning, PCR, and site-directed mutagenesis- E. coli expression and purification using Bio-rad FPLC- Enzyme kinetics using UV-vis spectroscopy and RP-HPLC- Isothermal titration calorimetryThe project was a joint project with Professor Gideon Davies, University of York and Professor Ben G. Davis, Oxford University and funded by BBSRC (Biotechnology and Biological Sciences Research Council)

The purpose of the research was to examine the interaction between two identified surface binding sites (which are not joined to the active site) in barley α-amylase 1 and starch. The experimental work included:- Site-directed mutagenesis - Protein expression in Pichia pastoris- Purification of barley α-amylase using Äkta explorer. - Enzyme kinetics using UV-vis spectroscopy and HPAE-PAD (Dionex). - Characterising starch binding using pull-down assays, confocal laser scanning microscopy and surface plasmon resonance (Biacore) I was responsible for teaching in the courses: “Biochemistry” (lectures) and “Experimental Biochemistry” (laboratory course). I was during my employment introduced to Vector NTI. The project was funded by the Carlsberg Foundation.
