The purpose of the research was to perform structure-function studies of retaining and inverting mannosyl transferases. In particular mannosylglycerate synthase from Rhodothermus marinus was used as a model system to study catalytic mechanisms of glycosyl transfer. The purpose of the research was to examine the interaction between two identified surface binding sites (which are not joined to the active site) in barley α-amylase 1 and starch. The experimental work included:- Site-directed mutagenesis - Protein expression in Pichia pastoris- Purification of barley α-amylase using Äkta explorer. - Enzyme kinetics using UV-vis spectroscopy and HPAE-PAD (Dionex). - Characterising starch binding using pull-down assays, confocal laser scanning microscopy and surface plasmon resonance (Biacore) I was responsible for teaching in the courses: “Biochemistry” (lectures) and “Experimental Biochemistry” (laboratory course). I was during my employment introduced to Vector NTI. The project was funded by the Carlsberg Foundation.
