Project I: • Designed and synthesized imidazoquinoline derivatives as modulators of Toll-like receptors (TLRs) 7 and 8. Characterized their impact on TLR induce NFκB expression in reporter cell lines and performed ELISA on compound-treated peripheral blood mononuclear cells (PBMCs) to monitor NFκB induced cytokine production. • Demonstrated a structural switch that turns agonists into antagonists, provided tools for facile antagonist development as potent autoimmune treatment.Project II• Constructed a library of molecules as Tumor Necrosis Factor Receptor 1 (TNFR1) antagonist and evaluated their potency in the disruption dimerization and the inhibition of NFkB activation. Demonstrated a structure-activity relationship and identified hit molecules and their potential as treatment of TNF induced rheumatoid arthritis.
