During the last 30 years, I have focused my research on various aspects of cancer research. Starting from my graduate school, I worked hand to hand with my mentor in designing a novel class of nitrosourea and phenothiazine containing compounds to target tumor cells, followed by post-doctoral work at the Indian Institute of Science, where I developed a mouse model to study apoptosis in vivo to better understand the role of thymidine/uracil in 5-Fluorouracil mediated cell death. During my second post-doctoral fellowship with Dr. Ted Lawrence, I developed a non-invasive imaging model to study the effect of tissue-specific gene delivery using adenoviral vectors to liver tumors. It was at this time I realized that drug-induced toxicity remains a major challenge in cancer therapy and I decided to investigate various aspects of cancer cell signaling to explore possibilities to target cancer cells more specifically. During the next 10 years, I studied EGFR signaling from cell-free systems to HNSCC patients' specimen collected before and during treatment to better understand the role of EGFR in cancer progression and effects on EGFR protein during treatment with chemotherapy, radiotherapy, or EGFR targeting agents. I continued to study EGFR with a primary focus on understanding how activated-EGFR protein is stabilized and using that information to develop new approaches to target activated EGFR protein to degradation. One of the agents that were developed during this process is going through IND enabling studies.
