My primary focus is to discover and delineate how the regulation of transcription influences erythroid gene expression, focusing on the expression of the globin genes. I work with human hematopoietic stem cells (HSCs), primary erythroid cultures, human umbilical cord-derived erythroid progenitors, and occasionally fresh bone marrow cells from donors. We use a combination of gene editing techniques (lentiviral transfection, CRISPR, lipofection, etc.) drug treatments, Fluorescence Assisted Cell Sorting (FACS), Flow Cytometry, DNA/RNA sequencing, etc. to probe our models of hematopoiesis.The average adult must produce ~2million red blood cells per second to maintain homeostasis, our focus is to understand the molecular mechanism that underly the development of red blood cells in both normal and disease states such as alpha and beta Thalassemia, Diamond-Blackfan Anemia, Sickle cell anemia, and other hemoglobinopathies.My end goal is to become proficient in the field of transcriptional regulation of hematopoiesis and translate my skills into a clinical setting and focus on the amelioration of symptoms of hemoglobinopathies.
