1. Finished Genotyping Five Populations of BC1F1 plants with KASP Markers for Four Domestication Mutants-DNA was extracted by first using a TissueLyzer machine and then a BioSprint machine-Quantified the DNA using a BioTek Epoch Microplate Spectrophotometer -Normalized the DNA-Created PCR plates and ran them through a Roche LightCycler 480 or Roche LightCycler 96-Identified the segregation ratio for each plant in each population for each domestication gene and the segregation ratio for the plants that were heterozygous for all four domestication gene mutants-Used Excel to run Chi-square tests to provide evidence for each plant population following the expected segregation ratio or to provide evidence that they were not following the expected segregation ratio2. Learned to Make Pennycress Crosses-How to emasculate the pennycress flowers (female parent) using a dissecting microscope-How to pollinate the emasculated flowers (male parent)3. Harvested Over 1,000 Pennycress Plants from our Greenhouses (with help from other undergraduates)-Using hand pans, removed the chaff and harvested seed was put into envelopes4. Cleaned Chaff from Yield Plant Trials Using a Clipper Office Tester Machine-Weighed the seed using a scale connected to a computer, which imputed the data into an Excel spreadsheet5. Hand Harvested Pennycress Field Plots, Including one in Morris, MN (with help from other undergraduates)-Also helped de-weed the plots 6. Labeled Harvested Pennycress Envelopes from the Greenhouse and the Tags for the Yield Plots7. Used the NIR (Near-infrared spectroscopy) Machine on Greenhouse Samples8. Used the MARViN ProLine Seed Analyzer Machine on Greenhouse Samples 9. Sprayed wheat scab (Fusarium Head Blight) Inoculum on Wheat Field Trials and Recorded the Scores after Infection (with help from other undergraduates)10. John Deere Gator Certification and use*Worked for Dr. Julia Zhang, UMN Department of Agronomy and Plant Genetics
