Director - Cmc Cell TherapyTriarm Therapeutics Jun 2024 - Present -
Associate Director - Cell TherapyBayer Dec 2021 - Jun 2024Leverkusen, North Rhine-Westphalia, De -
Senior Scientist - Process And Product DevelopmentAllogene Therapeutics Jul 2018 - Dec 2021South San Francisco, California, Us• Developed closed manufacturing scale Allogeneic CAR-T cell therapy process using automated cellprocessing equipment, working with vendors to evaluate new and existing technologies (Sefia, Sepax,LOVO, Ekko, Prodigy, Xuri, Erbi).• Oversaw process team production of CAR-T cells using transduction (LVV) and transfection(electroporation: MaxCyte, BTX, Nucleofector) to generate allogeneic CAR-T cells in bioreactor tomaximize therapeutic yield (CAR+%, KO%, TVC) while maintaining desired phenotypes.• Identified Drug Product manufacturing materials & staffing inefficiencies, enabling an 80%reduction in production run costs.• Spearheaded a new testing platform for manufacturing, utilizing emerging technologies toreduce development run costs by 98% per run.• Tech-Transferred development processes to CMO & internal manufacturing sites with batch recordgeneration/review, leading to the manufacture & clinical availability of treatments.• Worked with Quality Assurance and External Manufacturing-SC to address/resolve deviations, nonconformance issues and raw material supply during clinical manufacturing processes.• Managed pre-IND and early phase process projects, including personnel scheduling, process materialbudget of >$2M per year and ensuring materials and processes meet regulatory demands.• Led planning and execution of a $3M CAPEX project, doubling lab capacity and transitioning all work to a new site, ensuring timeline adherence and uninterrupted operations.• Created safety stock strategy for critical supply vulnerabilities, which kept all project timelines on trackand prevented work stoppage during COVID-19.• Managed staff, ensuring proper training and robust development plans, resulting in multiplestaff promotions. -
Postdoctoral Fellow - Continuous ManufacturingSanofi Genzyme 2016 - Jul 2018Paris, France, Fr• Led a new platform development project, increasing monoclonal antibody production by 250%& process robustness by 300%.• Developed strategy for $1.5M+ CAPEX purchase through vendor prototyping collaboration, reducing FTE & materials costs while increasing development throughput capabilities by 400%.• Created proof of concept process to link manufacturing unit operations resulting in thetransition from two manufacturing steps into a single step.• Scale up and scale down of perfusion based mammalian cell culture (CHO) for monoclonal antibody production from shake flasks to various scales of bioreactors including the use of single use (SUB) ambr® and WAVE bioreactors™ with a focus on high density perfusion processes.• Monitoring of metabolites and growth conditions (ViCell, Cedex Bio HT, RAPIDlab, osmometer, etc) -
Postdoctoral Research AssociateIowa State University May 2014 - Nov 2015Ames, Iowa, Us• Investigated a new treatment for Wilson’s Disease through translational medicine collaboration with the Helmholtz Center Munich in Germany• Created production process that increased output by 200%, meeting drug substance supplygoals for pre-clinical animal studies.• Solved critical study problem by creating a modified western blot immunoassay to test antibodies purified from rat serum using Protein G (affinity chromatography), due to its better binding of rat IgG than Protein A, in rats treated with a promising new treatment for Wilson’s disease• Preparative large-scale purification of extracellular proteins and secondary metabolites from fermentation broth for further study and use in human and animal medicine.• Scale up culture growth from agar plates to shake flasks to various sizes of bioreactors using controlled atmospheres.• Executed critical first stage of project for 4.5-million-dollar grant from Advanced Research Projects Agency-Energy (ARPA-E) funded program focused on conversion of natural gas to natural products for use in biofuels• Customized fed-batch fermentation process to maximize desired product concentration for generation of commercial product• Transferred process knowledge to commercial partner and supported initial technology implementation into their biofuel production plan • Membrane protein fractionation and purification under anaerobic conditions using Coy anaerobic chambers and using ion exchange (IEC) and size exclusion (SEC) chromatography with FPLC and AKTA.• Optimized cell lysis, osmotic shock, sonication, French press, high pressure homogenizer (Avestin EmulsiFlex-C3), in controlled atmosphere while monitoring enzyme stability and activity using gas chromatography (GC FID) assay and SDS-PAGE and western blots. -
National Science Foundation (Nsf) Gk-12 FellowIowa State University 2013 - 2014Ames, Iowa, Us• Mentored 210 students to improve critical reasoning skills, by creating engaging andinformative lesson plans.• Advocated for young people in underserved urban schools to gain an interest in Science,Technology, Engineering and Mathematics (STEM) fields through regular in class interactionwith science professionals. -
Research AssistantIowa State University Jun 2008 - 2014Ames, Iowa, Us• Regularly used several forms of spectroscopy (UV-Vis, Fluorescence, circular dichroism CD, atomic absorption AA) to monitor protein stability, structure, binding and aggregation.• Characterized a modified peptide methanobactin (mb) with one of the highest known binding constants, as high as 4.06 x 10^43 M-1, using isothermal titration calorimetry (ITC) Due to the limitations of the ITC and methanobactins extreme binding affinities an alternative displacement method had to be used resulting in a value of 1.47 x 10^24 M-1. Methanobactin’s high binding constants and ability to reduce several transition metals makes it of interest for many uses ranging from bioremediation to disease treatment. -
