Project: Hi-ChIPThe three-dimensional (3D) organization of chromatin in the nucleus gives a major impact on regulating gene expression. The invention of high-throughput chromosome conformation capture (Hi-C) technique has enabled us to observe genome architecture at high-resolution. On the other hand, deep sequencing is required to fully identify chromatin architectural features. Therefore, an improved method for mapping factor directed by chromatin conformation is needed. New techniques that combine Hi-C with chromatin immunoprecipitation-sequencing (ChIP-seq), called HiChIP showed significant improvement of regulatory (example: H3K27ac) and structural (example: cohesin) interactions with moderate sequencing depth. However, our ability to examine its biological function and role in disease is restricted by the large amounts of samples required to perform current experimental approaches. So now we are developing an improved HiChIP method for mapping factor of specific chromatin interaction maps using very low amounts of starting material (10^4 cells). For this purpose, I use mouse lens epithelial cells.
