Hands-on experience with a broad range of biochemical, cell-biological, immunological, microbiological, virological, and molecular-biological techniques including: - Biolayer interferometry (BLI, Sartorius, Octet) for screening purposes and to study antibody-antigen interactions (binding, competition, and affinity measurements).- Surface plasmon resonance (SPR, Cytiva, Biacore) for screening purposes and to study antibody-antigen interactions (binding, competition, and affinity measurements).- Flow cytometry (BD Biosciences FACS Canto II and other equipment) to analyze antibody-dependent cell-mediated cytotoxicity, complement-dependent cytotoxicity, interactions between antibodies and surface components of bacterial and mammalian cells and to detect proteins inside mammalian cells.- All kinds of enzyme-linked immunosorbent assay (ELISA) formats (design, development & qualification), radioimmunoassays & (immuno)cytology.- Bacterial diagnostics.- One-dimensional polyacrylamide gel electrophoresis of proteins & western blotting. -Mammalian cell culture & cell transfection, infection, and transduction (for protein production, adenovirus production or transgene expression).-Protein production, (affinity) purification, labeling, quantification and characterization (using various methods).- Adenovirus batch production (including plaque purification) and (CsCL) purification for research, HDEP and preGMP purposes including release assays, CofA, study plan and study report.- Virological assays: OD260, transgene expression & westernblot, VP-qPCR, QPA, ID PCR (identity and integrity), sanger sequencing results analysis (identity), productivity assay, reduced scale model productivity assay (Presence), transgene stability assay.- Molecular cloning: pUC to pAd by restriction enzyme digests & IVA cloning, bacterial transformation, mini & maxi culture & preps, restriction enzyme analysis, sanger sequence results analysis- Polymerase chain reactions (e.g. random amplification of polymorphic DNA, diagnostic, identity and integrity) & restriction fragment length polymorphism analysis- DNA agarose gel electrophoresis (including pulse-field gel electrophoresis) & Southern blotting.
