In this project, I unraveled a new corticostriatal circuit using optogenetics, electrophysiology, and pharmacology. This circuit is supported by a polysynaptic motif and conveys a second phase of excitation to striatal neurons through an acetylcholine-dependent glutamate release mechanism. These findings could have implications for behavior, plasticity, and learning. I developed a new circuit mapping method that combines optogenetics and multiple patch-clamp recordings. I found that connected pairs of neurons in the cerebral cortex share more inputs from the thalamus than unconnected pairs, supporting current models of cortical processing. The method I developed opened new possibilities for research in this area.
